Spinal kord
travmaları artan nüfus ile birlikte son yıllarda özellikle genç toplumda ciddi sağlık
problemi haline gelmiştir. Çalışmamızda spinal kord travmasını takiben uygulanan
metilprednizolon ve propolisin apopotozis üzerindeki etkilerini araştırmak amacıyla
dört deney grubu (n=10) oluşturulmuştur. İlk grup hariç tüm gruplarda, klip yöntemi
kullanılarak spinal travma modeli uygulanmıştır. Tartışmalar olsa da
metilprednizolon dışında etkin ve klinik kullanımda olan tedavi yöntemi
olmadığından dolayı deney gruplarında propolisin aktif bileşeni olan kafeik asit
fenilesterin etkilerini kıyaslamak amacıyla bir gruptaki sıçanlara metilprednizolon
tedavisi uygulanmıştır. Tedavi sonrasında 48. saatte tüm sıçanlar tekrar operasyona
alınmış ve histolojik inceleme için laminektomi sahasındaki spinal kord dokusu
çıkarılmıştır. Histolojik değerlendirme amacıyla Hematoksilen Eozin ve TUNEL
boyama yöntemleri kullanılmıştır. Hematoksilen Eozin boyama ile ışık mikroskobisi
altında hücrelerde nekroz, vakuolizasyon, hemoraji ve genel hasar değerlendirilirken
TUNEL yöntemi ile boyanan lamlarda ile ışık mikroskobi altında apopototik hücre
sayımı yapılmıştır. Sonuçta; genel hasar değerlendirildiğinde propolis kullanılan
grup, travma grubu ile kıyaslandığında metilprednizolona benzer olumlu sonuçlar
elde edilmiştir. Ayrıca apoptotik hücre sayımı gruplar arasında kıyaslandığında,
travma grubuna göre metilprednizolon ve propolis apopotozisi engellediği
görülmektedir. Genel hasardan farklı olarak propolisin antiapoptotik etkinliği
metilprednizolona kıyasla daha belirgin olduğu saptanmıştır. Bu bulgular ışığında,
insan vücuduna hiçbir zararı olmayan, propolisin aktif bileşeni olan kafeik asit
fenilesterin klinik kullanıma girebileceği fakat konu ile ilgili ayrıntılı, destekleyici
deneysel çalışmalara ihtiyaç olduğu düşünülmektedir.
Spinal cord injury has become a serious health problem with increased population in
the last years. In our study, rats were dividen four groups (n=10) to investigate the
effects of methylprednisolone and propolis in apopotosis following experimental
spinal cord injury. We applied aneurysm clip compression model all groups for
spinal cor injury except the first group. Through it is not clear, we used treatment of
methylprednisolone in one group for comparing the effects of caffeic acid phenethyl
ester which is the biologic effective ingredient of propolis. 48 hours after spinal cord
injury all rats operated again and spinal cord specimens taken for histologic
examination. The sections were stained with Hemotoxylin eosin and using the
apopotozis kit for TUNEL. Necrosis, vacuolization, hemorrhage and general damage
assesment with hemotoxylin eosin and apopototic cells count with TUNEL under the
light microscope. In conclusion, methylprednisolone and propolis effective in
supression of general damage as against the trauma group. Again, in the apopototic
cell count both methylprednisolone and propolis were effective than trauma group.
This study indicate that; caffeic acid phenethyl ester which is the biologic effective
ingredient of propolis may be use in clinical treatment of spinal cord injury but we
need some detailed and supported studies in this issue.
