Gelişim
dönemindeki stres maruziyeti, fetüsün beyin gelişimini olumsuz yönde etkileyerek
bireylerin yatkınlık derecelerine göre farklı psikopatolojik süreçlerin gelişimine
neden olabilmektedir. Kişiliği belirleyen ve davranışlarla ilgili hayati rol oynayan
merkezler prefrontal korteks (PFK)’te bulunmaktadır. Bu tez çalışmasında, erken
gelişim evrelerinden itibaren strese maruz kalan sıçanların PFK’deki nöronlarında
meydana değişiklikler tarafsız stereolojik sayım yöntemleriyle incelenmiş ve
yavruların emosyonel durumlarıyla olan ilişkisi araştırılmıştır. Prenatal ve postnatal
dönemlerde strese maruz bırakılan sıçanların anksiyete düzeyinde meydana gelen
değişiklikler ve depresyona yatkınlık düzeyleri değerlendirildikten sonra, nöronların
strese karşı verdikleri yanıtın derecesi c-Fos immunohistokimyası ile incelenmiştir.
Erken yaşam stresine maruz kalan dişi ve erkek sıçanların medial PFK’deki toplam
c-Fos (+) nöron sayıları kontrol gruplarına göre anlamlı düzeyde yüksek olarak
bulunmuştur. Erkek sıçanların anksiyete düzeyi ile toplam c-Fos (+) nöron sayısı
arasında pozitif bir korelasyon saptanırken, depresyona yatkınlık düzeyleri ile negatif
bir korelasyon saptanmıştır. Dişilerde ise anlamlı bir farklılık gözlenmemiştir. Golgi-
Cox yöntemiyle boyanan piramidal nöronların dendritik kompleksitesinde ve dikensi
çıkıntı yoğunluğunda da anlamlı yapısal değişiklikler gözlenmiştir. Stres maruziyeti,
apikal dendritler üzerindeki dikensi çıkıntıların morfolojik özelliklerinde, yavruların
anksiyete düzeyleri ile bağlantılı olarak değişikliklere neden olmuştur. Nöronlarda
gözlenen bu yapısal değişiklikler, sinaptik plastisite kapasitesini olumsuz etkileyerek
psikopatolojik bozuklukların gelişmesine zemin hazırlayabilir.
Exposure to stress
during development adversely affects the brain development of fetus and might lead
to the development of psychopathological processes that vary according to stress
predisposition of individuals. Centers playing a crucial role in determining an
individual's personality and behavior are found in the prefrontal cortex (PFC). In this
thesis study, changes occurring in neurons of the PFC of rats exposed to stress from
their early developmental stages were investigated by unbiased stereological
counting methods and their relation with the emotional status of the offspring was
examined. Following the evaluation of anxiety and depression susceptibility levels of
the offspring, response of neurons to stress was examined by using c-Fos
immunohistochemistry. In male and female rats exposed to early life stress, total
number of c-Fos (+) neurons in the medial PFC was significantly higher than those
of control groups. There was a positive correlation between the total number of c-Fos
(+) neurons and anxiety levels of male rats, whereas a negative correlation with the
susceptibility to depression levels. No significant differences were observed in
females. Significant structural changes were also observed in the dendritic
complexity and spine density of pyramidal neurons stained with Golgi-Cox method.
Depending on the anxiety level, early life stress exposure caused to changes in the
morphological properties of spiny protrusions on the apical dendrites. These
structural changes observed in neurons might predispose to the development of
psychopathological disorders by adversely affecting the synaptic plasticity capacity.
