Erişkin erkek sıçanlarda doksorubisin ile oluşturulan testis hasarı üzerine L-karnitinin etkisi

Abstract

Erişkin erkek sıçanlarda doksorubisin ile oluşturulan testis hasarı üzerine L-karnitinin etkisi. Doksorubisin, lösemi, lenfoma ve çeşitli kanser tiplerinde, dayanıklı tümörlere karşı antitümör ajanı olarak kullanılan antrasiklin tipi bir antibiyotiktir. Kanser tedavisinde kullanılan doksorubisin, kemoterapik bir ajandır. Sitotoksik etkilerine ek olarak çok sayıda ciddi yan etkileri de bulunmaktadır. L-karnitin, in vivo olarak metiyonin ve lizin aminoasitlerinden sentezlenen, antiapoptotik ve antioksidan etkileri kanıtlanmış vitamin benzeri bir amindir. Bu çalışmada antioksidan gibi davranan L-karnitinin doksorubisin ile hasara uğratılmış sıçan testis dokusuna olan etkisini histolojik olarak inceleyerek, L-karnitinin doksorubisine maruziyeti sonucu koruyucu etkisini incelemeyi amaçladık. Çalışmamızda toplam 28 adet erişkin Spraque-Dawley türü erkek sıçan kullanıldı. Sıçanlar her grupta 7 erişkin erkek sıçan olacak şekilde kontrol, 4 mg/kg doksorubisin, 200 mg/kg L-karnitin, 4 mg/kg doksorubisin+200 mg/kg L-karnitin verilen grup olmak üzere 4 gruba ayrıldı. Her hayvanın deney öncesi ve deney sonu vücut ağırlıkları ile deney sonunda testis ağırlıkları ölçülerek karşılaştırıldı. Sol testisler doku takip işlemi için Bouin çözeltisi içerisine, sağ testisler ise formaldehit çözeltisi içerisine alındı ve rutin histolojik işlemlerden sonra bloklandı. Elde edilen parafin bloklardan 3 μm kalınlığında seri kesitler alındı ve kesitler Hematoksilen+Eozin ve Periyodik Asit-Schiff+Hematoksilen ile boyanarak mikroskobik incelemeleri yapıldı. Çalışmanın sonucunda elde ettiğimiz bulgulara göre, doksorubisinin testis ve vücut ağırlığını azalttığı, testiste seminifer tübüllerde ve hücrelerde hasara yol açarak spermatogenezi etkilediği gözlendi. Doksorubisin ile birlikte verilen L-karnitinin ise testiste oluşan bu hasarı azaltabileceği gözlendi.

Effects of L-carnitine on testicular toxicity induced by doxorubicin in adult male rats. Doxorubicin is an anthracycline antibiotic used as a antitumor agent against the resolute tumors in leucemia, lymphoma, other cancer types and is used. Doxorubicin which is used for cancer treatments is chemotherapeutic agent that has lots of side effects especially on the testicular tissue. L-carnitine, is a vitamine like amine synthesized from methionine and lysine amino acids in vivo, of which antiapoptotic and antioxidant effects has been proved. In this study, to investigate histological effects of L-carnitine which is acted as an antioxidant, testicular tissue of rats that are damaged by using doxorubicin are studied. We aim to investigate protective effect of L-carnitine against the exposure of doxorubicin. A total of 28 Spraque-Dawley male rats were investigated on this research. Rats were divided into 4 groups having 7 adult rats in each. Consisting of; control group; 4 mg/kg doxorubicin group; 200 mg/kg L-carnitine group; 4 mg/kg doxorubicin + 200 mg/kg L-carnitine group. Body and testicle weights of each rats were before and after experiments. Also weight of testicle measured after experiments. Left testicle was placed into Bouin solution; and right testicle was placed into formaldehyde solution before bracketing for routine histological process. Sections which are 3 μm thick were taken from paraffine blocks and stained with Hematoxylene+ Eosin; Acid- Schiff+ Hematoxylene and then sections were investigated in light microscope. Results showed that doxorubicin cause the loss in rats body weight and testicle weight. A doxorubicin also effected the spermatogenesis by damaging the seminiferous tubule and cells. On the other hand, administration of L-carnitine together with doxorubicin might lead to reduction in damage level observed in testicle.
