Aroma, yani koku ve tat maddeleri gıda, içecek, kozmetik, deterjan, kimya ve
ilaç sektörlerinde yaygın kullanım alanı olan katkı maddeleridir. Genelde doğal
kaynaklardan veya kimyasal sentez yoluyla üretilmektedir. Aroma maddelerinin
bitkilerden saflaştırılmaları genelde pahalı bir süreçtir. Kimyasal sentez ise yaygın
kullanılan iyi bir alternatif olmakla birlikte kimyasal ürünlerin insan sağlığını ve çevreyi
tehdit etmesi ve tüketicilerin doğal ürünleri tercih etmeleri, kullanımlarını
azaltmaktadır. Bu durum çevreye uyumlu ve doğal ürün üretimine yönelik talebi
artırmış ve son yıllarda biyoteknolojik yöntemlerin kullanımı önem kazanmıştır.
Biyoteknolojik yöntemlerde elde edilen aroma maddesi üretimi, günümüzde sadece
düşük düzeyde gerçekleşmektedir.
Bu çalışmada terpen yapısında olan α-iyonol bileşiğinin 19 farklı
mikroorganizma ile ön denemeleri yapıldı. Bu çalışmalar sonucunda α-İyonolün A.
Niger ve F. culmarum ile biyotransformasyonları sonucu oluşan metabolit
karışımlarının aroma özelliği gösterdiği belirlendi. Söz konusu biyotransformasyon
tepkimeleri preparatif ölçekte tekrarlandı. Elde edilen metabolitler kolon kromatografisi
ile saflaştırıldı ve yapıları GK-KS, 1D ve 2D NMR teknikleri ile aydınlatıldı. Her iki
biyotransformasyon sonucu verimleri % 6,6 (A. niger) ve % 15,3 (F. culmarum) olan
aynı metabolit elde edildi.
Aroma compounds are flavour and fragrance which are highly important
ingredients commonly used in many industries such as food, beverage, cosmetics,
detergents and pharmaceuticals. Most of these compounds are produced by either
chemical synthesis or extraction from plant sources. Obtaining aroma compounds from
plants is expensive due to their isolation difficulties. Chemical synthesis serve as a good
alternative for the production of aroma substances however they are not preferred due to
harmful effects to the health and environment and the consumer’s preference for natural
products. This urged the need for the development of new environmentally friendly
processes. In recent years, the utilisation of biotechnological processes for the
production of aroma compounds appeared to be an important choice, however, still low
percentages of are derived from bioprocesses.
In this study, pre-biotransformation of a terpene, α-ionol, was carried out by 19
different microorganisms. As a result it was found out that exract obtained from
biotransformation of α-ionol with A. niger and F. culmorum showed aroma properties.
These biotransformation reactions were repeated on preparative scale. The metabolites
were purified by column chromatography and their structures were elucidated by using
GC-MS, 1D and 2D NMR spectra. Both biotransformation reactions produced the same
metabolite with % 6,6 (A. niger) and % 15,3 (F. culmorum) yield, respectively.
