Nanopartikül sentezi için çevre dostu ve düşük maliyetli yöntemlere duyulan ihtiyaç dolayısıyla biyolojik (yeşil sentez) yaklaşımlara ilgi artmaktadır. Nanopartiküllerin yeşil sentezinde bitkiler, funguslar ve bakterilerin kullanıldığı çok sayıda çalışmalar yapılmıştırr. Her organizmanın avantajı ve dezavantajı bulunmakta fakat, funguslar ile biyokütle uygulamalarının basitliği, hücre içi metal emilimi ve hızlı büyümeleri ve çok çeşitli hücre içi ve dışı enzim üretmeleri nanopartikül biyosentezinde onları öne çıkarmaktadır.
Bu çalışmada hipersalin ortamdan izole edilen Aspergilus flavus, A. clavatus ve A. fumigatus biyokütleleri kullanılarak gümüş nanopartiküllerin (AgNP) biyosentez koşulları Box-Behnken Deney tasarımı ile optimize edilmiştir. Sentezlenen AgNP’ler UV-vis spektroskopisi, Dinamik ışık saçılımı (DLS), Zeta potansiyeli, Transmisyon Elektron Mikroskobu (TEM) ve X-ışını kırınımı (XRD) kullanılarak karakterize edildi. Antimikrobiyal aktivitesi disk difüzyon yöntemi ile antoksidan aktivitesi ise DPPH ile değerlendirilmiştir.
PDI, zeta potansiyeli ve partikül boyutu verilerine göre yapılan deney tasarımı analizi, AgNP sentezinde en verimli koşulların A. flavus için 27℃/9pH/0,1mM, 27℃/9pH/0,5mM ve 37℃/9pH/0,3mM, A.fumigatus için 27℃/9pH/0,1mM ve 37℃/7pH/0,5mM olduğu belirlenmiştir. UV-vis spektroskopisinde yapılan ölçümlerde AgNP'ler, 420nm ile 430nm arasında karakteristik yüzey plazmon rezonansı göstermiştir. A. flavus ve A. fumigatus ile sentezlenen AgNP’ler 50-100nm arasında değişen, küresel morfolojiye sahipti. Kullanılan test organizmaları E. coli, S. aureus ve C. albicans ın sentezlenen Ag nanopartiküllere duyarlı olduğu belirlenmiştir. Yapılan optimizasyon ve karakterizasyon sonucunda fungusların yeşil sentez ile AgNP üretiminde potansiyel araçlar olabileceği sonucuna varılmıştır
There is increasing interest in biological (green synthesis) approaches due to the need for environmentally friendly and low-cost methods for nanoparticle synthesis. Numerous studies have been conducted using plants, fungi and bacteria in the green synthesis of nanoparticles. Each organism has advantages and disadvantages, but the simplicity of biomass applications with fungi, their intracellular metal absorption and rapid growth, and the production of a wide variety of intracellular and extracellular enzymes make them stand out in nanoparticle biosynthesis.
In this study, biosynthesis conditions of silver nanoparticles (AgNP) were optimized by Box-Behnken Experiment design by using Aspergilus flavus, A. clavatus and A. fumigatus biomass isolated from hypersaline medium. The synthesized AgNPs were characterized using UV-vis spectroscopy, Dynamic light scattering (DLS), Zeta potential, Transmission Electron Microscopy (TEM), X-ray diffraction (XRD) and. Its antimicrobial activity and antioxidant activity were evaluated by disc diffusion method and DPPH, respectively.
Experimental design analysis based on PDI, zeta potential and particle size data showed that the most efficient conditions for AgNP synthesis were 27℃/9pH/0.1mM, 27℃/9pH/0.5mM, and 37℃/9pH/0.3mM for A. flavus. It was determined that 27℃/9pH/0.1mM and 37℃/7pH/0.5mM for A.fumigatus. In measurements made in UV-vis spectroscopy, AgNPs showed characteristic surface plasmon resonance between 420nm and 430nm. AgNPs synthesized by A. flavus and A. fumigatus had spherical morphology ranging from 50-100nm. It was determined that the used test organisms E. coli, S. aureus and C. albicans were sensitive to the synthesized Ag nanoparticles. As a result of the optimization and characterization, it was concluded that fungi can be potential tools in the production of AgNP by green synthesis
