Anadolu bal arısı olarak bilinen Apis mellifera anatoliaca, Türkiye’nin kuzeydoğu
ve güneydoğu bölgeleri hariç tüm orta ve kıyı kesimlerinde çok geniş yayılışa sahip olan
bir ırktır. Bal arısı venomu uzun yıllardır geleneksel tıp uygulamaları ile tedavi amaçlı
kullanılıyor olmasına karşın bal arısı venomlarının bileşenleri ile gerçekleştirilen
biyokimyasal çalışmalar ırklar arasındaki farkları/benzerlikleri, moleküler çalışmalar ise
bileşenlerin biyolojik etkilerinin etki mekanizmasını detaylı olarak ortaya koyacak nitelikte
değildir.
Bu çalışma kapsamında Apis mellifera anatoliaca venomu bileşenleri
kromatografik ve elektroforetik yöntemler ile ayrıştırılarak, tüm dünyada ve Türkiye’de
kadınlar arasında görülme sıklığı ilk sırada yer alan meme kanseri tedavisinde
kullanılabilirliğinin değerlendirilmesi amaçlanmıştır. Bu amaçla majör venom peptid
bileşenlerinin sitotoksik ve anti-proliferatif etkinlikleri in vitro yöntemler ile taranmıştır.
Ana 11 adı verilen bileşenin anti-proliferatif, anti-invasiv, apoptotik ve hücre döngüsü
üzerindeki etkisi belirlenerek bu etkinin mekanizması transkripsiyonel ve translasyonel
seviyede araştırılmıştır. Etkin peptidin moleküler ağırlığının 2846,45Da ve birincil
yapısının GIGAVLKVLTTGLPALISWIKRKRQQ-NH2 olduğu kütle spektrometresi ve
amino asit dizilemesi teknikleri ile ortaya çıkarılmıştır. Dizinin biyolojik veri
tabanlarındaki karşılaştırmalı analizi ile bu peptidin melittin olduğu ve çalışma boyunca
elde edilen kromatografik verilere göre Apis mellifera anatoliaca liyofilize venomu
içindeki oranı %58,1 olarak belirlenmiştir.
Sunulan tez çalışmasının sonucunda ülkemiz doğal zenginlikleri arasında yer alan
Apis mellifera anatoliaca venomunun in vitro anti-kanser ve anti-metastatik etkinliğinden
sorumlu peptid bileşeni tanımlanmıştır. Apis mellifera anatoliaca kaynaklı melittinin
çalışma kapsamında belirlenen anti-kanser ve anti-metastatik etkinliğinin in vivo
çalışmalarla tedavi amaçlı kullanım için araştırılması çalışma sonuçlarının
sürdürülebilirliğine katkı sağlayacaktır
Apis mellifera anatoliaca as known as the Anatolian honey bee, is a race that has a
very wide distribution in all central and coastal parts of Turkey, except for the northeastern
and southeastern regions. Although honey bee venom has been used for therapeutic
purposes for many years with traditional medicine applications, biochemical studies with
components of honey bee venoms do not reveal the differences/similarities between races,
and molecular studies do not reveal the mechanism of action of the biological effects of the
components in detail.
Within the scope of this study, it was aimed to evaluate the utility of Apis mellifera
anatoliaca venom components, by separating with chromatographic and electrophoretic
methods, in the treatment of breast cancer, the incidence of which is the first among
women in the world and also in Turkey. For this purpose, cytotoxic and anti-proliferative
effects of major venom peptide components were screened by in vitro methods. The antiproliferative,
anti-invasive, apoptotic and cell cycle effects of the component Ana 11 was
determined and the mechanisms of these effects were investigated at the transcriptional
and translational level. The molecular weight of the effective peptide was revealed as
2846.45Da and the primary structure was GIGAVLKVLTTGLPALISWIKRKRQQ-NH2,
by mass spectrometric and amino acid sequencing techniques. By comparative analysis of
the sequence in biological databases, this peptide was determined as melittin and its ratio
in lyophilized venom of Apis mellifera anatoliaca was determined as 58.1% according to
the chromatographic data obtained throughout the study.
As a result of the presented thesis the peptide component responsible for the in
vitro anti-cancer and anti-metastatic activities of Apis mellifera anatoliaca venom, which
is among the natural resources of our country, was identified. Investigating the anti-cancer
and anti-metastatic activities of melittin isolated from Apis mellifera anatoliaca,
determined within the scope of the study, for therapeutic use with in vivo studies will
contribute to the sustainability of the results of this study
