Filamentli funguslar çeşitli enzimleri yüksek miktarlarda üretim yetenekleri ile ön
plana çıkmaktadır. Bununla birlikte, halotolerant/halofilik fungusların enzimleri sahip
oldukları özellikler bakımından çeşitli avantajlar sunmaktadır.
Bu çalışmada hipersalin ortamlardan izole edilen mikrofungus izolatlarının α-amilaz
enzimi aktivitelerine göre taranması, üretim koşullarının optimizasyonu, elde edilen enzimin
kısmi karakterizasyonu ve derin kültür fermentör uygulamasıyla üretimi amaçlanmıştır.
Hipersalin ortamdan izole edilen kırk adet mikrofungus izolatı amilolotik
aktivitelerine göre kalitatif olarak belirlenmiştir. %5, %7, ve %12 ve %17 tuz
konsantrasyonu içeren nişasta agar besiyerinde en iyi aktivite gösteren fungusların
Aspergillus clavatus, A. tamarii ve A. sydowii olduğu gözlemlenmiştir. Bu üç mikrofungus
türünden A. tamarii α-amilaz üreticisi olarak seçilmiştir. Üretim koşullarının
optimizasyonunda, tuz, pH ve inokulum biyokütle miktarını etkileyen faktörler olarak
bulunmuştur. Optimum koşullarda gerçekleştirilen derin kültür fermentör uygulaması ile
enzim aktivitesi 403 U/ml olarak bulunmuştur. Üretilen α-amilaz’ın kısmi karakterizasyonu
ile, 55 ℃ - 60 ℃ sıcaklık aralığında 30 dakika inkübasyon süresince 1,40 – 1,60 mg /ml
nişasta konsantrasyonunda, pH 6.0’ da ve 2 M tuz konsantrasyonunda optimum aktivite
göstermiştir.
Bu çalışma hipersalin çevrelerdeki fungal çeşitliliğin amilolitik aktiviteye sahip
üretici mikrofungusları içerdiğini ve bu ortama adapte olan funguslar tarafından üretilen
amilazların tuza toleranslarının yüksek olduğunu göstermektedir. α-amilaz üreticisi olarak
A. tamarii pellet morfolojisinin üretim için uygun olması, tuz varlığında gelişim ve
enzimatik aktivite göstermesi ile öne çıkan fungustur
Filamentous fungi stand out with their ability to produce various enzymes in high
amounts. However, the enzymes of halotolerant / halophilic fungi offer various advantages
in terms of their properties.
In this study, it was aimed to screen microfungus isolates isolated from hypersaline
enviroments according to α-amylase enzyme activities, optimization of production
conditions, partial characterization of the enzyme and production by submerged fermenter
application.
Forty microfungus isolates isolated from hypersaline media were qualitatively
determined according to their amylolotic activity. Aspergillus clavatus, A. tamarii and A.
sydowii were the fungi that showed the best activity on starch agar medium containing 5%,
7%, 12% and 17% salt concentrations. From these three microfungus species, A. tamarii was
chosen as the α-amylase producer. In optimization of production conditions, salt, pH and
inoculum were found to be factors affecting the amount of biomass. Enzyme activity was
found to be 403 U / ml with submerged culture fermenter application performed under
optimum conditions. With the partial characterization of the α-amylase produced, it showed
optimum activity at 1,40 – 1,60 mg/ml starch concentration, pH 6.0 and 2 M salt
concentration during 30 minutes incubation between 55 ℃- 60 ℃ temperature.
This study shows that fungal diversity in hypersaline environments includes
productive microfungi with amylolytic activity and amylases produced by fungi that adapt
to this environment have high salt tolerance. As a producer of α-amylase, A. tamarii is a
prominent fungus with its pellet morphology suitable for production, growth in the presence
of salt, and enzymatic activity
