Çalışmanın amacı, 5-florourasil uygulaması ile oluşan gastrointestinal mukoza hasarının önlenmesi ve/veya tedavi edilmesinde berberinin kullanım potansiyelinin araştırılmasıdır.
Yöntem: Deney kapsamında hayvanlar modele uygun olarak 5 gruba ayrılmıştır. Kontrol grubundaki hayvanlara sadece oral yoldan fosfat tamponu uygulanırken; 5FU, BER75, BER150 ve BER300 gruplarında yer alan hayvanlara 7 gün boyunca sırasıyla gün aşırı intraperitonal enjeksiyonla 5-florourasil uygulanmış, diğer günlerde ise yanak içine travma uygulanmıştır. BER75, BER150 ve BER300 gruplarında yer alan hayvanlara sırasıyla 75, 150 ve 300 mg/kg dozlarda berberin oral yoldan uygulanmıştır. Deneyin başında ve sonunda hayvanların vücut ağırlıkları değerlendirilmek üzere ölçülmüştür. Hayvanların kalplerinden kan alınmış ve ELISA kitleri kullanılarak total antioksidan aktivite (TAS), total oksidan aktivite (TOS), TNF-α, IL-6 ve NF-κB düzeyleri ölçülmüştür.
Bulgular: TAS ve TOS düzeylerinden hareketle grupların oksidatif stres indeks değerleri hesaplanmış ve gruplar arasında anlamlı fark bulunamamıştır. Berberin uygulanan gruplarda ölçülen TNF-α, IL-6 ve NF-κB düzeyleri sadece 5-FU uygulanan gruba kıyasla anlamlı derecede azalmıştır. Ötenazi sonrasında jejenum ve ileum dokularından numune alınarak histopatolojik değerlendirme yapılmıştır. Değerlendirmede 5-florourasilin yarattığı doku hasarının izleri gözlemlenmiştir.
Tartışma ve sonuç: Vücut ağırlıklarındaki değişim oranları, sitokin düzeylerindeki değişimler ve histolojik değerlendirmeye ait bulgular bir arada değerlendirildiğinde elde edilen sonuçların literatür ile uyumlu olduğu görülmektedir. Tez çalışmasının sonucunda berberinin doza bağımlı olarak 5-florourasilin neden olduğu gastrointestinal sistemdeki hasarın önlenmesinde ve/veya tedavisinde etkili olabileceğini söylemek mümkündür
In our study, the main purpose is to investigate berberine’s potential use in prevention and/or treatment of mucosal damage in gastrointestinal system induced by 5-fluorouracil treatment.
Method: Animals were separated to 5 groups, and the groups were determined according to the experiment model. For 7 days, while animals in control group were applied only phosphate buffer orally; animals in 5FU, BER75, BER150 and BER300 groups were applied 5-fluorouracil intraperitoneally or traumatized in the inner lip tissue every other day. Animals in BER75, BER150 and BER300 groups were applied 75, 150 and 300 mg/kg berberine, every day for 7 days. At the beginning and the end of the trial, animals’ weights were measured in order to be evaluated later. Blood was drawn from hearts of the animals at the end of the experiments and (total antioxidant status (TAS), total oxidant status (TOS), TNF-α, IL-6 and NF-κB levels were measured using ELISA kits.
Results: Experimental groups’ oxidative stress indexes were calculated using TAS and TOS levels, and no significant difference was found. TNF-α, IL-6 and NF-κB proinflammatory cytokines levels of animals in groups that have berberine in their dose regimens were decreased significantly compared to the group treated with 5-fluorouracil only. After euthanasia, samples were collected from jejunum and ileum tissues and evaluated histopathologically. Tissue damage caused by 5-fluorouracil was observed during histopathological evaluation.
Discussion and conclusion: When the changes of body weight and cytokine levels were evaluated with findings of histopathological examination, obtained results are consistent with the literature. At the end of this thesis, it is possible to say that berberine may be effective in the treatment and/or prophylaxis of the damage in the gastrointestinal system caused by 5-fluorouracil in a dose-dependent manner
