C3H5NO kimyasal formülüyle bilinen akrilamit beyaz renge sahip suda
çok iyi çözünebilen bir monomerdir. Birçok endüstriyel alanda kullanılmasının
yanında akrilamit ayrıca yüksek sıcaklığa maruz kalan karbonhidratça zengin
yiyeceklerde de oluşmaktadır. Hayvan ve insanlarda yapılan farklı
farmakolojik ve epidemiyolojik çalışmalarda akrilamitin nörotoksik etkiler
gösterdiği bildirilmiştir. Timokinon çörek otunun aktif maddesidir.
Timokinonun anti-inflamatuar, antikonvülsan, antitüsif, anti-tümör ve
antioksidan aktiviteleri de içeren birçok faydalı özelliği bulunmaktadır.
Kuersetin ise çeşitli yiyeceklerde bulunan bir flavonoittir. Antioksidan, anti inflamatuar ve antiviral etkilere sahiptir. C6 hücre hattı sıçan glioblastoma
hücreleridir. Literatürde akrilamitin C6 hücreleri üzerindeki etkilerini
gösteren bir araştırmaya rastlamadık. Ancak, önceki laboratuvar
deneyimlerimiz bize akrilamitin C6 hücreleri üzerinde toksik etkilerinin
olabileceğini göstermiştir. Bu çalışmamızda, akrilamitin C6 hücreleri
üzerindeki etkilerinin belirlenmesi ve timokinon ile kuersetinin akrilamitin
etkisini ne yönde etkilediğinin araştırılması amaçlanmıştır.
Çalışmamızda ilk olarak MTT testi ile akrilamitin 24 saat için C6
glioblastoma hücreleri için toksik dozu bulundu. Sonra timokinon ve
kuersetinin 24 saat için C6 hücrelerine toksik olmayan dozları MTT
kolorimetrik testi kullanılarak belirlendi. Daha sonra bulunan bu dozlar
kullanılarak apoptoz yolağına bakmak için akış sitometrisi ile kaspaz 3/7 ve
anneksin V testleri yapıldı. Ayrıca otofajik ölüm ve oksidatif stres de akış
sitometrisi analiziyle değerlendirildi. İmmünositokimyasal olarak Nrf2
miktarı incelendi. Morfolojik inceleme TEM ve konfokal mikroskop
analizleriyle yapıldı.
Bulgularımıza bakıldığında C6 hücrelerinde toksik etki oluşturmak için 4
mM akrilamit değeri belirlendi. Timokinon ve kuersetinin toksik olmayan
dozları sırasıyla 3.9 ve 2.0 µM olarak belirlendi. Timokinon ve kuersetinin
anneksin V ve kaspaz 3/7 sonuçlarına bakıldığında akrilamitin neden olduğu
apoptozu ve mikroskopik incelemelerde de morfolojik deformasyonları azalttığı

görüldü. Otofaji incelemesinde ise akrilamitin neden olduğu otofajinin
timokinon ve kuersetin verilmesiyle azaldığı ortaya konuldu. Nrf2 ifadesine
bakıldığında akrilamitin Nrf2 ifadesini arttırdığı, timokinon ve kuersetinin
verilmesinin bu ifadeyi daha da arttırdığı gözlemlendi.
Sonuç olarak, akrilamitin C6 glioblastoma hücreleri üzerinde hasar
yapıcı etkisinin bulunduğu, timokinon ve kuersetinin önceden verilmesinin bu
hücrelerde akrilamit kaynaklı hasara karşı koruyucu etkilerinin olduğu
saptanmıştır. Timokinon ve kuersetinin diğer akrilamit ilişkili hücre, doku
veya organ hasarlarında da koruyucu etki gösterme potansiyeline sahip
olduğuna ve ileriki çalışmalarda denenebileceğine inanıyoruz
Acrylamide, known as the formula of C3H5NO, is a white-colored and
readily water-soluble monomer. Besides being used in many industrial areas,
acrylamide is also formed in carbohydrate-rich foods processed at high
temperatures. Acrylamide was reported to exhibit neurotoxic effects in
different pharmacological and epidemiological studies conducted on animals
and humans. Thymoquinone is the active constituent of Nigella sativa and has
many beneficial properties, including anti-inflammatory, anticonvulsant,
antitussive, anti-tumor and antioxidant activities. Quercetin is a flavonoid
found in various foods. It has antioxidant, anti-viral and anti-inflammatory
properties. C6 cells are rat glioblastoma cells. In the literature, to the best of
our knowledge, there is not any study showing the effects of acrylamide on C6
cells in literature. However, our previous laboratory experience showed that
acrylamide exerts toxic effects in C6 cells. In this study, we aimed at
attempting to prevent the toxicity of acrylamide against C6 cells by using
thymoquinone and quercetin.
In our study, first, the toxic dose of acrylamide against C6 glioblastoma
cells for 24 hours was determined by MTT test. Then, non-toxic doses of
thymokinone and quercetin against C6 cells for 24 hours were determined by
MTT colorimetric test. After that, using these doses, caspase 3/7 and annexin
V tests were performed by flow cytometry to evaluate whether apoptosis
pathway was inducted. Furthermore, autophagy and oxidative stress were also
assessed by flow cytometry. The amount of Nrf2 was immunocytochemically
examined. Morphological examination was performed by TEM and confocal
microscope analyses.
As a result, the 4 mM of acrylamide was determined to be used to induce
toxicity in C6 cells. The non-toxic doses of thymokinone and quercetin were
respectively determined as 3.9 ve 2.0 µM. Thymokinone and quercetin not only
reduced both acrylamide-induced apoptosis in annexin V and caspase 3/7
results and morpholocial deformations in microscopic examinations. In
autophagy, it was revealed that acrylamide-induced autophagy was decreased

by quercetin and thymokinone pretreatments. As for Nrf2 expression, it was
observed that acrylamide increased Nrf2 expression and thymkinone and
quercetin pretreatments increased this expression even further.
In conclusion, in the study, acrylamide had a damaging effect on C6
glioblastoma cells and thymokinone and quercetin pretreatments exerted a
protective effect against acrylamide-induced damage in C6 cells. We believe
thymokinone and quercetin might be used in future studies due to their
potential to exhibit protective effects against other acrylamide-related cell and
organ damages
