Bu çalışmada DPPH radikal süpürücü aktiviteleri ile gerçekleştirilen tarama sonrası
seçilen 4 Ganoderma izolatının kültür sıvısı ekstraksiyonu ile elde edilen ekstraktların
antioksidan, antimikrobiyal ve sitotoksik etkisi araştırılarak değerlendirilmiştir.
Antioksidan aktivitenin araştırılması amacı ile DPPH radikal süpürücü aktivite,
redükleyici güç, metal şelatlayıcı aktivite ve beta-karoten linoleik asit yöntemi
uygulanmıştır. OBCC 6069 kodlu izolat ekstraktının DPPH süpürücü aktivite yönteminde
1,934 mg/ml, redükleyici güç 0,416 mg/ml ve metal şelatlayıcı aktivite yönteminde 2,730
mg/ml EC50 değerine sahip olduğu belirlenmiştir. Antimikrobiyal aktivitenin araştırılması
için disk difüzyon yöntemi uygulanmış ve Pseudomonas aeruginosa (ATCC 27853),
Escherichia coli (ATCC 25922) suşlarına karşı antimikrobiyal etki gözlemlenmiştir. MİK
ve MBK deneyleri uygulanarak etkin dozlara ulaşılmıştır. MTT yöntemi ile izolat
ekstraktının sitotoksik etkisinin araştırılması ardından, OBCC 3 ve OBCC 5109 kodlu
izolatların apoptotik etki varlıkları sorgulanmıştır
In the study, it was investigated antioxidant, antimicrobial and cytotoxic effects of 4
Ganoderma isolates which were selected a screening program depends on their DPPH
radical scavenging activities.
DPPH radical scavenging activity, reducing power, metal chelating activity and beta carotene linoleic acid method have been applied for the investigation of antioxidant activity.
The OBCC 6069 coded isolate extract was determined to have 1.934, 0.41 and 2.730 mg/ml
EC50 values for DPPH radical scavenging, reducing power and metal chelating activities,
respectively. To determine antimicrobial activity, disc disfussion method was applied and
antimicrobial effect was observed against Pseudomonas aeruginosa (ATCC 27853),
Escherichia coli (ATCC 25922) strains. Effective doses have been obtained by applying
MIC and MBC experiments. After determining of the cytotoxic effect of isolate extracts with
MTT assay, the presence of apoptotic effect of OBCC 3 and OBCC 5109 coded isolates was
researched
