Serbest radikaller ve antioksidanlarla ilgili yapılan çalışmaların önemi her geçen gün daha fazla artmaktadır. Normal şartlarda canlı metabolizmasında serbest radikaller devamlı olarak üretilmektedir ve bu du rum antioksidanlarla denge halindedir. Ancak bazı durumlarda bu denge serbest radikaller lehine değişir. Böyle durumlarda organizma serbest radikallerin vücutta meydana getirdiği hasara karşı savunma yapabilmek için antioksidanlara ihtiyaç duyar. Antioksid anların en önemli özelliklerinden biri serbest radikallerin neden olduğu oksidatif stresi önlemektir. Son yıllarda antioksidan etkileri bakımından fenolik bileşikler dikkat çekmektedir. Bu çalışmada kullandığımız kumarik asit bir hidroksisinamik asit türe vi olan fenolik bileşiklerdendir. Yapılan bu tez çalışmamızda kumarik asitin ovaryan kanser hücreleri üzerindeki etkilerini inceledik. Yöntem: Hücre kültürü çalışmamızda OVCAR OVCAR-3 hücre hattı kullanıldı. RPMI (Roswell Park Memorial Institute) besiyerler inin içine %10 FBS ve %1 penisilin streptomisin eklenerek OVCAR OVCAR-3 hücreleri çoğaltıldı. Değişen dozlarda kumarik asit uygulaması sonrası kumarik asitin hücreler üzerine etkileri, sitotoksisite testleri (MTT, Nötral red), inverted mikroskop incelemeleri (He matoksilen Eozin boyama, immunohistokimyasal boyama) ve yara iyileşmesi testi ile araştırıldı. Bulgular: Yaptığımız deneylerde kumarik asitin artan dozlarında hücre canlılığının azaldığı, çekirdek ve sitoplazma morfolojisinin bozulduğu, hücre migrasyonunun yavaşladığı, apoptotik yolaklarda görev alan proteinlerin ifadesinin arttığı tespit edildi. Sonuç: Sonuç olarak kumarik asitin over kanseri hücre serileri olan OVCAR OVCAR-3 hücrelerinde sitotoksik ve antimigratuar etkilerini ortaya koymuş olup, tümör hücreleri nde apopitozisi arttırdığını gözlemledik
The importance of studies on free radicals and antioxidants is increasing
day by day. Under normal conditions, free radicals are constantly produced in l iving
metabolism and are in balance with these antioxidants. However, in some cases, this
balance shifts in favor of free radicals. In such cases, the organism needs antioxidants to
defend against the damage caused by free radicals in the body. One of the most
important properties of antioxidants is to prevent oxidative stress caused by free radicals.
In recent years, phenolic compounds have attracted much attention in terms of
antioxidant effects. The coumaric acid we used in the study is one of the phenol ic
compounds, a hydroxycinnamic acid derivative. In our experiments, we examined the
effects of coumaric acid on ovarian cancer cells.
Method: We used the OVCAR OVCAR-3 cell line in our cell culture study. OVCAR OVCAR-3 cells were grown in the medium obtained by addi ng 10% FBS and 1% penicillin
streptomycin into RPMI (Roswell Park Memorial Institute) media. We investigated the
effect on cells after application of varying doses of coumaric acid by cytotoxicity tests
(MTT, Neutral red), inverted microscope examinations (Hematoxylin Hematoxylin-Eosin staining,
immunohistochemical staining), and wound healing test.
Results: In our experiments, it was determined that increasing doses of
coumaric acid decreased cell viability, deteriorated the morphology of the nucleus and
cytoplasm, s lowed down cell migration, and increased the expression of proteins
involved in apoptotic pathways.
Conclusion: As a result, we observed that coumaric acid showed cytotoxic and
antimigratory effects in OVCAR OVCAR-3 cells, which are ovarian cancer cell lines, and
increased apoptosis in tumor cells.
