Bu ça lışma, Ocak 2018-Ocak 2020 tarihleri arasında 75 hafif-orta düzeyde ROP gelişen an cak tedavisiz vaskülarizasyonu tamamlanan olgu kontrol grubu olarak, takipleri esna sında tedavi uygulanan 73 ciddi ROP olgusu tedavi grubu olarak çalışmamıza alındı.
Genetik analiz için kan alınıp VEGF-A geninde 3 bölge, HIF-2α geninde 3 bölge,
BDNF geninde 3 bölge, NOS geninde 2 bölge olmak üzere toplam 11 SNPs bölgesinin
analiz sonuçları iki grup arasında karşılaştırıldı. İki grup doğum kilosu, doğum haftası,
sistemik durumları, kaç gün yoğun bakım ünitesinde tedavi gördükleri, yoğun bakımda
kaldıkları sürelerde gördükleri tedaviler açısından da karşılaştırıldı. 11 polimorfizm
bölgesinden 8’inde istatistiksel olarak anlamlı sonuçlar elde ettik. Bu bölgelerden dör dünde (rs2010963, rs3025039, rs13419896, rs2070744), mutant alellerin fazlalığının,
ROP şiddetini artırarak olguların tedavi endikasyonu alması açısından risk artışına ne den olduğunu gösterdik (p<0,001, p<0,001, p=0,022, p=0,004). Diğer dört bölgede ise
(rs833061, rs7929344, rs1867785, rs1868085) ise mutant alellerin varlığında, ROP
kliniğinin daha hafif seyrederek, tedavi gerekmeden, kendiliğinden sonlanma ihtima linin arttığını gösterdik (p<0,001, p=0,019, p=0,017, p=0,017). Sonuç olarak çevresel
ve demografik faktörlerin yanında genetik analiz sonuçlarımıza göre genetik yatkınlı ğın, ROP kliniği ve seyri üzerinde etkisi olabileceğini düşünüyoruz. Çalışmamızdaki
rs2010963, rs3025039, rs13419896, rs2070744 polimorfizmlerinin, hastalık şiddeti ile
ilişkili olabileceğini gösterdik
This study inclu ded 75 mild to moderate ROP between January 2018 and January 2020 but completed
vascularization without treatment, and 73 severe ROP cases treated during their fol low-up were included in our study. For genetic analysis, blood samples were taken and
3 regions in the VEGF-A gene, 3 regions in the HIF-2α gene, 3 regions in the BDNF
gene and 2 regions in the NOS gene were analyzed between the two groups. The two
groups were compared in terms of birth weight, birth week, systemic conditions, how
many days they were treated in the intensive care unit and the treatments they received
during their stay in the intensive care unit. We obtained statistically significant results
in 8 of the 11 polymorphism regions. In four of these regions (rs2010963, rs3025039,
rs13419896, rs2070744), we showed that the excess of mutant aleles increased the
ROP severity, causing an increased risk for patients to receive treatment indications (p
<0.001, p <0.001, p = 0.022, p = 0.004). In the other four regions (rs833061,
rs7929344, rs1867785, rs1868085), we showed that ROP spontaneously regressed wit hout treatment in the presence of mutant aleles (p <0.001, p = 0.019, p = 0.017, p =
0.017). As a result, we think that genetic predisposition may have an impact on ROP
severity according to our genetic analysis results as well as environmental and demog raphic factors. We have shown that the polymorphisms of rs2010963, rs3025039,
rs13419896, rs2070744 in our study may be related to disease severity
