Klotho geni birçok biyolojik süreçte yer alan anti-aging, anti-inflamatuvar ve anti-tümör özellikleri olan humoral bir faktördür. Klotho geninin ifadesindeki azalma, kolon kanseri de dahil olmak üzere çoğu kanser tipinde malign oluşumlara neden olur. Klothonun antitümör etkisi, kanser hücrelerinde malign oluşuma neden olan sinyal yolaklarını inhibe etmesinden kaynaklanır. Tümör nekroz faktörüne bağlı apoptozu indükleyen ligand (TRAIL), kanser hücrelerinde apoptozu indükleyen tümör nekroz faktör (TNF) sitokin süper ailesinin bir üyesidir. İnsan tümörlerinin %60 gibi önemli bir oranı TRAIL'e karşı direnç gösterir. TRAIL, kanser tedavisinde kemoterapötik toksisiteye neden olmadan kanser hücrelerinin proliferasyonunu azaltarak apoptozu indükler. TRAIL, ölüm reseptörleri olan DR4 ve DR5 ile etkileşime girerek apoptotik etki gösterir.
Bu çalışmadaki amaç, TRAIL dirençli insan adenokarsinom kolon kanseri hücrelerinde (Caco-2), apoptoz indükleyicisi olduğu bilinen klotho geninin ekspresyonunu değiştirerek (gen aktivasyonu ve knock-out), bu ekspresyon değişimlerinin kanser hücrelerindeki TRAIL aracılı apoptotik duyarlılığa etkisinin olup olmadığını araştırmaktı. Böylece klotho geninin önemli fonksiyonlarından biri olan apoptotik sürecin, ilk kez TRAIL ölüm reseptörleri (DR4, DR5, DcR1 ve DcR2) üzerinden analiz edilmesi sağlandı. Bu amaçla öncelikle klotho geni CRISPR/Cas9 tekniği ile Caco-2 hücrelerinde klotho geni aktifleştirildi ve knock-out edildi. Klotho geninde yapılan bu ekspresyon değişikliklerinin kanser hücrelerindeki etkisi; annexinV/PI flow sitometri, WST-8, qRT-PCR, invazyon, koloni oluşturma potansiyeli ve immunohistokimyasal analizler ile değerlendirildi.
Sonuçlarımızda Caco-2 hücrelerinde klotho gen ekspresyon artışının hücrelerin proliferasyon, invazyon ve koloni oluşturma potasiyelini baskıladığı ve kaspaz aktivitesini artırdığı görüldü. Bu etkinin gen aktivasyonuna bağlı olup olmadığı ise genin knock-out edilmesiyle test edildi. Bulgularımız klotho gen aktivasyonunun apoptoza dirençli kanser hücresinde antitümör etkiye sahip olduğunu gösterdi.

The klotho gene is a humoral factor with anti-aging, antiinflammatory and antitumor properties, which is involved in many biological processes. Decreased expression of the klotho gene causes malignant formations in most types of cancer, including colon cancer. Klotho's antitumor effect is due to the inhibition of signalling pathways that cause malignant formation in cancer cells. Tumor necrosis factor-induced apoptosis inducing ligand (TRAIL) is a member of the TNF cytokine superfamily that induces apoptosis in cancer cells. A significant 60% of human tumors show resistance to TRAIL. TRAIL induces apoptosis in cancer treatment by reducing the proliferation of cancer cells without causing chemotherapeutic toxicity. TRAIL interacts with the death receptors DR4 and DR5 and demonstrate an apoptotic effect.
The aim of this study was to investigate whether these expression changes have an effect on TRAIL-mediated apoptotic sensitivity in cancer cells by altering (overexpression and knock-out) the expression of the klotho gene, which is an apoptosis inducer, in TRAIL-resistant human adenocarcinoma colon cancer cells (Caco-2). Thus, the apoptotic process, one of the important functions of the klotho gene, was analysed for the first time on TRAIL death receptors (DR4, DR5, DcR1 and DcR2). For that purpose, the klotho gene was firstly klotho gene activation and knock-out on Caco-2 cells using the CRISPR/Cas9 technique. The effect of these expression changes in the klotho gene on cancer cells; annexin V/PI flow cytometry, WST-8, qRT-PCR, invasion and colony-forming potential and immunohistochemical analysis.
In our results, it was seen that the increase in klotho gene expression in Caco-2 cells inhibited the proliferation, invasion and colony forming potential of the cells and increased the caspase activity. It was determined that the decrease in klotho gene expression had no effect on the viability of Caco-2 cells. These results show that klotho gene activation has an antitumor effect in cancer cells resistant to apoptosis. vivo studies and that klotho will be an important basis for basic cancer studies in order to be a strong marker in the treatment of colon cancer
