Ülseratif kolitte karaciğer dokusunun hasarlandığı bilinmektedir. Kolit nedenli
karaciğer hasarını önlemek için birçok sentetik ve doğal maddeler kullanılmasına rağmen
henüz tam bir başarıya ulaşılamamıştır. Bu çalışmada sıçanlarda deneysel olarak 120 mg/kg
TNBS verilerek oluşturulan kolitte gelişen oksidatif stres ve karaciğer hasarına karşı L NAME ve likopenin koruyucu etkileri araştırıldı. Çalışmada 105 adet 220-250 gr ağırlığında
Sprague Dawley ırkı erkek sıçanlar kullanıldı. Sıçanlar her grupta 7 hayvan olacak şekilde
kontrol grubu hariç 12 gruba ayrıldı. Kontrol grubuna 1 ml serum fizyolojik intraperitonal
(i.p) verildi. Diğer tüm gruplara sıfırıncı (0.) günde %50 etenolde çözülerek hazırlanmış
TNBS intrarektal olarak verilip ülseratif kolit oluşturuldu. 1. 2. ve 3. gruplar TNBS grupları
olarak belirlendi. TNBS uygulamasından 1 gün sonra 4. 5. ve 6. gruptaki hayvanlara L NAME, 7. 8. ve 9. gruptaki hayvanlara 1 mg/kg zeytinyağı, 10. 11 ve 12. gruptaki hayvanlara
ise 5 mg/kg likopen i.p olarak verildi. Sadece TNBS verilen hayvanlar 1. 2. ve 3. günlerde
diğer gruplardaki hayvanlar ise 2. 3. ve 4. günlerde eter ile anestezi edilerek kan ve karaciğer
doku örnekleri alındı. Alınan kandan serum ALT, AST ve LDH seviyeleri ölçüldü. Karaciğer
dokularında rutin histolojik doku takibi yapılarak mikroskobik inceleme yapıldı. Serum ALT,
AST ve LDH değerleri sadece 120 mg/kg TNBS verilen gruplarda oldukça artmış ve bu
durum kolitin karaciğer dokusuna verdiği hasarı çok ciddi bir biçimde yansıtmıştır. Bu
gruplarda biyokimyasal bulgular histolojik bulgularla da desteklenmiştir. Nitekim karaciğer
dokusunda ödem, sinüzoidal dilatasyon, nekroz, vakolizasyon, iltihabi hücrelerin birikimi gibi
doku hasarı parametreleri gözlenmiştir.
Deneysel sonuçlarımız hem serum AST, ALT ve LDH düzeyleri göz önüne alındığında
hem de karaciğer doku örneklerindeki histopatolojik bulguları değerlendirildiğinde likopenin,
kolit nedenli karaciğer hasarını önlemede zeytinyağından ve sentetik bir antioksidan olan L NAME’ den daha yararlı olduğunu ortaya koymuştur. Ayrıca kolitte karaciğer hasarını
gidermede likopen kullanıldığına dair bir literatüre rastlanmadı. Bu nedenle çalışmamız, kolit
nedenli karaciğer hasarında likopenin koruyucu etkisini araştıran özgün bir çalışmadır.
Ulcerative colitis is known to cause damage to the liver tissue. Although synthetic and
natural substances have been used to prevent liver damage due to colitis, little success has
been achieved so far. The present study experimentally induced colitis in rats given TNBS
(120 mg/kg) to investigate protective effect of L-NAME and lycopene upon oxidative stress
and liver damage in colitis. 105 male rats belonging in the species of Sprague Dawley, whose
weights varied between 220 and 250, were used in this study. These rats were divided into 12
groups, each consisting of 7 rats, except for the control group. 1ml of serum physiologic was
administrated intraperitoneally (i.p.) to the rats in the control group, while all the other groups
were given TNBS dissolved in %50 ethanol through the intrarectal tract on the very first day
of the experiment. Groups 1, 2 and 3 were labelled as TNBS groups. 24 hours after the
administration of TNBS, rats in groups 4, 5 and 6 were given L-NAME, the ones in groups 7,
8, 9 being given 1ml/kg olive oil. The remaining groups (10, 11 and 12) were given 5 mg/kg
of lycopene i.p. Those given only TNBS were anaesthetized with ether for blood and liver
tissue samples on the first, second and third days of the experiment, while those in the other
groups underwent the same application on the second, third and fourth days. Serum ALT,
AST and LDH levels obtained from the blood samples of the rats were analysed under the
microscope through a routine histological monitoring of the liver tissues. These values
increased to significant levels only in the groups exposed to TNBS, which reflects severity of
the damage to the liver tissue induced by colitis. Our biochemical findings were supported by
the histological ones. Also, parameters of tissue damage like oedema, sinusoidal dilatation,
necrosis, vacuolisation and accumulation of inflammatory cells were observed in the liver
tissue.
Evaluation of serum AST, ALT and LDH levels, as well as histopathological findings
of the liver tissue, seems to suggest that lycopene has a protective effect upon liver damage
due to colitis, and that lycopene is more beneficial to preventing oxidative stress than are
olive oil and L-NAME, an synthetic antioxidant substance. We could find no published study
into the protective effect of lycopene on colitis-induced liver damage. Therefore, our study is
the first of its kind in that we showed lycopene to have a protective effect upon colitis-related
liver damage.
