Sprague Dawley female rats were randomly divided into eight equal groups (n=7).
All groups were classified by their reperfussion times. Four groups were named as
‘1. groups’ and four groups were named as ‘2. groups’. After disection of right
inguinal region at all subjects of the experiment; femoral artery was clamped and
ischemia was performed at 4 hours. After ischemic period, right femoral artery was
canuled with 39 gauge ensulin syringe. Ġn control groups, only ischemia and
reperfusion was done. Ġn M1 and M2 groups, at the dose of 3 mg/kg %25 mannitol
solution, in HA1 and HA2 groups at the volume of 0.5 cc Human Albumin solution
and in EC1 and EC2 groups at the volume of 0.5 cc Euro Collins Solution was used as
reperfusion solutions to perfuse ischemic right lower extremity before reperfussion
period begans. At 1. groups, 4 hour reperfusion was done. At 2. groups, after
perfusing ischemic extremity with the perfusion solutions used as the same as 1.
group; soft tissues and skin was sutured and one week reperfusion was done. After
reperfusion period ended in all groups, soleus muscle biopsies from right extremity
and lung biopsies were taken for histological reserch. Samples were stained with
hematoksilen-eosin and examined by light microscopy and scored according to the
parameters described before. Results were analysed statistically and comparing
control groups with the groups at which perfusion solutions was used; the results
were statistically meaningful. As a result, regardig to our study; before reperfusion,
perfusing ischemic tissue by its own artery with the solutions which have antioxidant
properties, reduces ischemia/reperfusion injury.

56 adet Sprague Dawley cinsi dişi rat randomize olarak eşit sayıda (n=7), 1. Grup ve 2. Grup olmak üzere iki ana grup ve dört alt gruplara ayrıldı. Tüm gruplarda; sağ inguinal bölge diseksiyonunun ardından sağ femoral arter klemplenerek, sağ alt ekstremite 4 saat iskemiye bırakıldı. Ġskemi süresinin ardından, femoral arter 39 gauge’lik insülin enjektörüyle kanülize edildi. Alt gruplardan Kontrol gruplarına herhangi bir solüsyon kullanılmadı. Reperfüzyon öncesi M1 gruplarına 3 mg/kg dozda %25’lik mannitol solüsyonu ile; HA1 gruplarına 0.5 cc Human Albumin solüsyonu ile; EC1 gruplarına 0.5 cc Euro Collins Solüsyonu ile intravasküler olarak, iskemik sağ alt ekstremite perfüzyonu uygulandı. 1. gruplarda , 4 saatlik reperfüzyon yapıldı. 2. gruplarda; iskemik ekstremite için, 1. gruplarda kullanılan perfüzyon solüsyonları, reperfüzyon öncesi aynı dozda ve aynı yolla kullanılarak, yumuşak dokular ve cilt sütüre edildi ve sağ alt ekstremite bir hafta reperfüzyona bırakıldı. Tüm gruplarda reperfüzyon süresinin ardından, histolojik incelemeler için soleus kası ve akciğer örnekleri alındı. Histolojik incelemede, dokular hematoksilen-eosin boyasıyla boyanıp ışık mikroskobunda değerlendirildi. Belirlenen parametreler için histolojik skorlamalar yapıldı. Sonuçlar, istatistiksel olarak değerlendirildiğinde; 1. gruplarda bulunan ile 2. gruplarda bulunan kontrol grupları ile perfüzyon solüsyonlarının uygulandığı gruplar arasında ileri derecede farklılık mevcuttu. Bu çalışmanın sonucu; iskemik olan dokunun intravasküler yolla reperfüzyon öncesi, antioksidan özelliği olan solüsyonlarla perfüzyonunun yapılması iskemi/reperfüzyon hasarını azaltmaktadır.
