Cinsel saldırı mağdurlarının su ile temasının sperm tespit
edilebilirliği ve prostat spesifik antijen(PSA) aktivitesi üzerine olumsuz etkilerinin
olabileceği bilinmektedir. Temas edilen suyun özellikleri (kimyasal, biyolojik vs.) de
önem arz etmektedir. Bu çalışmada, in vitro ortamda farklı sularda sperm tespiti ve
PSA etkinliği üzerine araştırılması amaçlanmıştır. Bu çalışmada gönüllü ve
aydınlatılmış onamı alınan 30 farklı genç yetişkin erkekten cinsel perhiz sonrasında
alınan ejakülatlar farklı sıvı ortamlara konularak sperm hücreleri ve PSA
pozitifliğinin zamana bağlı değişimleri incelenmiştir. Her biri mililitresinde 50
milyon sperm hücresi içerecek şekilde standardize edilen semen örnekleri, deniz
suyu, çeşme suyu, distile su ve yöremizden geçen Porsuk Nehri’nin suyunu içeren 4
adet tüpte %20 oranında dilue edilmiştir. Dilue örneklerden günlük periyotlarla
sperm sayımı yapılmıştır. Yine aynı miktarda ve farklı sular içeren 4 adet tüpe eşit
hacimde ejakülat örneği konulmuştur. Aynı hacimde sperm içeren tüplerde 1/104
,
1/105 ve 1/(5x105
) oranlarında dilusyon işlemi yapılarak günlük periyotlarla PSA
etkinlikleri değerlendirilmiştir. Sperm hücrelerinin çeşme suyu ve distile suda deniz
suyu ve tatlı suya göre daha uzun süre tespit edilebildikleri saptanmıştır. Çeşme
suyunda 1128.saate, deniz suyunda 888.saate, distile suda 1008.saate ve porsuk
suyunda 792.saate kadar sperm hücresi görülebilmiştir. PSA etkinliğinin ise 1/104
dilusyonda; çeşme suyunda 1296.saate, distile suda 1008.saate, deniz suyunda
240.saate porsuk suyunda 264.saate kadar tespit edilebildiği, 1/105 ve 1/(5x105
)
oranlarındaki dilusyonlarda da çeşme suyu ve distile sularda tespit edilebilen PSA
etkinliğinin, deniz ve porsuk sularında tespit edilen süreden uzun olduğu
görülmüştür. İncelenen örneklerde; aradan geçen süre, dilüsyon oranı, sıvının
çeşidine göre sperm sayısında ne gibi değişikliklerin olduğu kaydedilmiştir. İn vitro
ortamdaki sonuçların in vivo ortam sonuçları ile aynı olmayacağı bilinmekle birlikte
değişik su örneklerinde sperm hücresi görülme ve PSA etkinliğiyle ilgili zamana
bağlı değişimler tablolar ve şekiller halinde sunulmuştur.
It’s known that victims
of sexual assault, contact with water, sperm and prostate-specific antigen(PSA)
detection may have negative effects contact the water properties (chemical,
biological, etc..) is also important. The aim of this study is to investigate in vitro
settings the sperm detection and the effect of prostate specific antigen (PSA)
presence at different liquids. In this study, volunteers received and informed consent
30 different young adults, after sexual abstinence ejaculates by putting the sperm
cells different liquid environments, and PSA changes over time to was examined. in
each milliliter will contain 50 million sperm cells in semen samples standartizated
and it was dilueted 20% with sea water, fountain water, distilled water and Porsuk
River water from our region in 4 tubes. Sperm count with daily periods of dilue
examples were made by sperm counting chamber. Equal volume samples of
ejaculate were put into 4 tubes that containing the same amount of different waters.
Dilution has been done rates of 1/104
, 1/105 and 1/5x105 in the tubes containing the
same volume of sperm and PSA activity was evaluated with daily period. Sperm
cells were detected in the fountain water and distilled water longer than sea water
and fresh water. The sperm cell could be seen in fountain water until at 1128 hour,
in sea water until at 888 hour, in distilled water until at 1008 hour, in fresh water
until at 792 hour. PSA activity was detected at 1/104 dilution, in fountain water until
at 1296 hour, in sea water until at 240 hour, in distilled water until at 1104 hour, in
