Vücutta meydana gelen doku hasarı veya patojen mikroorganizma
enfeksiyonundan sonra sistemik veya bölgesel yanıtlar oluşur. Hasara karşı oluşan bu
cevaba inflamatuar yanıt veya inflamasyon denir. İnflamasyon; sepsis, aterosklerotik
hastalıklar, kanser, romatoid artrit ve myokard infarktüsü gibi birçok hastalığın
patofizyolojisinde önemli rol oynar.
Lipopolisakkarit (LPS), gram negatif bakterilerin dış zarının bir parçası olup,
inflamatuar sitokinlerin, serbest oksijen radikallerinin, nitrik oksit ve araşidonik asit
metabolitlerinin aşırı üretimine yol açarak çeşitli inflamatuar hastalıkların
patogenezinde önemli rol oynar.
Propolis, çeşitli biyolojik ve farmakolojik özelliklerinden dolayı son yıllarda
araştırmacıların ilgisini çeken doğal bir bal arısı ürünüdür. Antibakteriyel, antioksidan,
antiviral, anti-inflamatuar, antifungal, antitümör, antihepatotoksik, immünomodülatör,
lokal anestezik gibi birçok etkiye sahiptir.
Bu çalışmada propolis ekstraktının deneysel inflamasyon ve antioksidan sistem
üzerine olan etkisinin araştırılması amaçlanmıştır.
Bunun için lipopolisakkarit (LPS) ile oluşturulan deneysel inflamasyon modeli
kullanıldı. Çalışmamızda 42 adet 3 aylık Sprague Dawley soyu erkek sıçan ile 6 deney
grubu oluşturuldu. Üç gruba intraperitoneal olarak 1 mg/kg LPS (E. coli, serotip 055-
B5) uygulandı. LPS enjeksiyonundan 24 saat sonra tedavi gruplarına 30 ve 90 mg/kg
propolis ekstraktı gavaj yolu ile verildi. FDG-PET taraması ile inflamasyon durumunu
belirlenmek için PET taramasından 1 saat önce ketamin anestezisi altında 18F -fluoro deoxy- D-glucose (0,8 ml/kg) intrakardiyak olarak uygulandı. FDG-PET analizinden
sonra kan ve doku örnekleri toplandı. Akciğer ve karaciğer 18F-FDG tutulumu
hesaplandı. Karaciğer, böbrek, akciğer dokuları ve eritrosit malondialdehit (MDA)
düzeyi, katalaz (KAT) ve süperoksit dismutaz (SOD) enzim aktivitesi belirlendi.
Çalışma sonunda akciğer, böbrek ve hemolizat MDA seviyeleri inflamasyon
kontrol grubunda yüksek bulunurken propolis ile tedavi edilen inflamasyon gruplarında
düşük bulundu. İnflamasyon kontrol gruplarının SOD aktivitesi akciğer dokusu ve
hemolizatta düşük bulunurken propolis ile tedavi edilen inflamasyon gruplarında yüksek bulundu. Benzer şekilde inflamasyon kontrol gruplarının KAT seviyesi akciğer dokusu
ve hemolizatta düşük bulunurken propolis ile tedavi edilen inflamasyon gruplarında
yüksek bulundu. Akciğer ve karaciğer 18F-FDG tutulumu inflamasyon kontrol
gruplarında yüksek olup propolis ile tedavi edilen inflamasyon gruplarında düşük
bulundu.
Sonuç olarak, propolisin sıçanlarda LPS ile oluşturulan inflamasyonun ve serbest
radikallerin azaltılmasında etkili olduğu bulunmuştur.
After tissue injury or infection by pathogen microorganisms in the body,
responses of systemic and local events are triggered. This generalized response to injury
is referred to inflammatory response or inflammation. Inflammation plays key role in
the pathophysiology of many diseases such as sepsis, atherosclerotic disease, cancer,
rheumatoid arthritis and myocardial infarction.
Lipopolysaccharide (LPS) is a component of the outer membrane of gram negative bacteria and it has an important role in the pathogenesis of several
inflammatory diseases by causing excessive release of inflammatory cytokines, oxygen
free radicals, nitric oxide, arachidonic acid metabolites.
Propolis is a natural bee-product, and because of several biological and
pharmacological properties it has attracted researchers’ interest in recent years. It has
many effects including antibacterial, antioxidant, antiviral, anti-inflammatory,
antifungal, antitumor, antihepatotoxic, immunomodulatory, local anesthetic and
anticancer activity.
The aim of this study was to investigate the effects of propolis extract on
experimental inflammation and antioxidant system.
We used the experimental lipopolysaccharide (LPS)-induced model. Totally, there
were 42, 4-month old male Sprague Dawley rats in 6 groups in the study design. In
three groups, 1mg/kg of LPS (E. coli, serotype055-B5) was administered to the rats
intraperitoneally. 30 mg/kg and 90 mg/kg of propolis extracts were given orally to
treatment groups after 24-hours of intraperitoneal LPS injection.
To determine the lung and liver inflammation status via FDG-PET, 18F-fluoro deoxy-D-glucose (0,8ml/kg) was administrated under the anesthesia by intracardiac
injection before the 1h of PET scanning. After the FDG-PET analyses, blood and tissue
samples were collected. Lung and liver 18F-FDG uptake was calculated. MDA, SOD,
and CAT levels were measured in tissues and erythrocyte.
In the study, MDA levels of hemolysate, lung and liver tissues were increased in
untreated inflammation group whereas decreased in inflammation groups treated with
propolis. SOD activities of inflammation groups were found to be decreased in lung tissue and hemolysate whereas SOD activities of propolis treated inflammation groups
were increased. Similarly, CAT activities of inflammation groups were found to be
decreased in lung tissue and hemolysate whereas CAT activities of propolis treated
inflammation groups were increased.
Lung and liver 18F-FDG uptake of inflammation groups were increased while lung
and liver 18F-FDG uptake of propolis treated inflammation groups were decreased.
As a result, propolis was found to be efficient in reducing inflammation and free
oxygen radical production induced by LPS in rats.
