Bu çalışmada, selüloz adsorbanının bazı diyabet (Akarboz (D1), Metformin (D2), Gliklazid (D3)), kolesterol (Gemfibrozil (K1), Ezetimib (K2), Fenofibrat (K3)) ve tansiyon (Atenolol (T1), Metoprolol (T2), Karvedilol (T3)) ilaçları ile adsorpsiyon kabiliyetinin ikili ve üçlü ilaç etkileşimleri araştırılmıştır.
Teorik hesaplamalarda MOPAC2012 paket programında PM6 ve PM7, Gaussian 09 paket programında DFT hibrit yaklaşımı, adsorpsiyon deneylerinde ilaç aktif maddelerinin selüloz üzerindeki adsorpsiyonu ve kontrollü salımı UV-Görünür Alan Spektrofotometresiyle incelenmiştir. Mopac2012 yönteminde vücut sıcaklığında (T=310K) kan fazında belirlenen K2-T2, K2-T3, K1-D2, K2-D1-T1, K3-D3-T2, K3-D2, K1-D2-T3 ve K1-D2-T3 molekülleri, DFT yönteminde K1-D2, K3-D1, K1-D2-T1 ve K3-D1-T3 molekülleri kullanıldığında birbirlerinin etkisini azaltacağı ya da artıracağı için mutlaka hekim tarafından doz ayarlaması yapılarak kullanılması gerekmektedir. Deneysel çalışmalarda ise vücut sıcaklığında pH=3‟te K1-T2 ve K1-D2-T2 moleküllerinin en yüksek salım yüzdesine sahip moleküller olduğu belirlenmiştir. Salım yüzdesinin yüksek olması ilaç molekülleri arasında etkileşimin olmadığını göstermektedir. Yani aralarında etkileşim olmayan K1-T2 ve K1-D2-T2 ilaç molekülleri selüloza adsorbe edilerek kullanıldığında ilaç vücutta daha uzun süre kalacak ve tedavi edici etkisini daha uzun süre gösterecektir.
Yapılan çalışmaların sonuçlarında etkileşimin termodinamik yönde gerçekleşmesi ve olası H-bağları için uyum halinde olması Mopac2012 ve DFT yöntemlerinde paralellik göstermektedir. Her iki yöntemde de etkileşim kararlılığı düşük olarak belirlenen K1-T2 ve K1-D2-T2 moleküllerinin deneysel çalışmalarda da en yüksek salım yüzdesine sahip olması uygulanan yöntemlerin sonuçlarının uyum içinde olduğunu göstermektedir.
In this study, adsorption ability of cellulose as adsorbent was investigated with binary and ternary drug interactions by using some diabetes (Acarbose (D1), Metformine (D2), Gliclazide (D3)), anti-cholesterol (Gemfibrozil (K1), Ezetimibe (K2), Fenofibrate (K3)) and anti- tension drugs (Atenolol (T1), Metoprolol (T2), Carvedilol (T3)).
Adsorption of drug active ingredients on cellulose and controlled release were investigated by using PM6 and PM7 methods of MOPAC2012 package program and DFT hybride approach of Gaussian 09 package program in theoretical calculations, and also by UV-Visible Spectrophotometer in adsorption experiments. According to theoretical calculations, the values obtained for blood phase at body temperature (T=310K) by Mopac2012 method, K2-T2, K2-T3, K1-D2, K2-D1-T1, K3-D3-T2, K3-D2, K1-D2-T3 and K1-D2-T3 molecules, and by DFT method, K1-D2, K3-D1, K1-D2-T1 and K3-D1-T3 molecules showed properties of increasing or decreasing effect on each other, so they necessarily have to be used with dosing by doctors. However in experimental studies in body temperature at pH=3 it was determined that, the molecules of K1-T2 and K1-D2-T2 have the maximum release percent. The higher release percents indicate the lack of interactions between the drug molecules. Thus when K1-T2 and K1-D2-T2 drug molecules which have no interaction among them are used by being adsorbed on cellulose, the drug will exist for a long time in body and its therapeutic effect will last longer. In the results of the study, the thermodynamic nature of the interactions results and being in tune for possible H-bonds showed parallelism in Mopac2012 and DFT methods. Both methods indicate that K1-T2 and K1-D2-T2 molecules have low interaction stabilities and also in experimental study the same molecules showed the highest percentage of release. This means being in concert with the results of the methods used.
