İşitme kaybı önemli algılama bozukluklarından biridir. Konjenital işitme kaybı yaklaşık her 1000 çocukta 1 görülür ve bunun yaklaşık yarısının genetik etyolojisi mevcuttur. GJB2 geni işitme kaybının genetik temelinde etkin olan en önemli genlerden biridir.
MLPA yöntemi delesyon, amplifikasyon ve nokta mutasyonları tek reaksiyonla
inceleyebilen bir yöntemdir. Salsa MLPA P163 B1 prob kiti kullanılarak GJB2, GJB3 ve GJB6 genleri incelenmiştir. Çalışmamızda orta, ileri ve çok ileri düzey nonsendromik nörosensöriyal tip işitme kaybı olan 90 olgu MLPA yöntemi ile incelenmiştir. GJB2 genindeki 35delG mutasyonu 90 olgunun 22 (%24.4) sinde homozigot, 6 sında (%6.6) heterozigot olarak saptanmıştır. GJB3 ve GJB6 genlerinde mutasyon saptanmamıştır. GJB2 geni için saptamış olduğumuz mutasyon oranları literatür ile uyumlu bulunmuştur. İşitme kaybının ailesel olması ve işitme kaybının
düzeyinin ileri derece olması 35delG mutasyon görülme oranını arttıran etmenler olmuştur. Çalışmamızda MLPA yöntemi non-sendromik nörosensöriyal tip işitme kaybı olan bireylerde başarı ile uygulanmıştır. Populasyonumuzda sık görülen mutasyonların
da MLPA prob dizaynı yapılarak yöntemin uygulanması ile daha iyi sonuçlar elde
Hearing loss is an important problem about sensation. Congenital hearing loss is
observed in 1 of 1000 children end half of this has a genetic etiology. GJB2 gene is one
of the most important genes taking part in genetic basis of hearing loss. MLPA
technique has the power to analyze deletions, amplifications and point mutations in a
single assay. GJB2, GJB3 and GJB6 genes were analyzed using Salsa MLPA P163 B1
probe set. In our study, 90 cases with modarate, severe and profound level non
syndromic neurosensorial type hearing loss were examined with MLPA technique.
35delG mutation in GJB2 gene was observed in 22 of 90 (24.4%) cases as homozygous
and 6 of 90 (6.6%) as heterozygous. Mutation was not observed in GJB3 and GJB6.
Mutation frequency of GJB2 in our study is comparable with the literature. Familial
hearing loss and advanced hearing loss are the factors that increasing the frequency of
35delG mutation. In our study, MLPA technique was applied to non-syndromic
neurosensorial type hearing loss cases successfully. Better results may be obtained by
applying the technique with designing MLPA probes for frequent mutations in our
population
