Çevresel stres faktörleri, tek başına bitkilerin verimlerini ve besin değerlerini etkiler,
ayrıca bitki patojenlerinin habitatlarını da olumsuz yönde etkileyerek patojenlerin
yayılmasına neden olur. Bu nedenle, bitkiler aynı anda farklı stres faktörlerine maruz
kaldıkları yerde yaşamak için çeşitli stratejiler geliştirmelidir. Bu tez çalışmasında, tuz
(NaCl) stresi, Rhizoctonia solani ve bu iki stres faktörünün kombinasyonuna farklı tepkiler
gösteren patates çeşitlerinin fizyolojik özelliklerinde ve sukroz metabolizmasında meydana
gelen değişikliklerin moleküler ve gen ifadesi düzeyinde belirlenmesi hedeflenmiştir. Bu
amaçla, üç aşama olarak planlanan denemede 19 ticari patates çeşidi, fizyolojik ve
morfolojik farklılıklar ile verim ve verim öğeleri vasıtasıyla ilk olarak tuzluluk toleransı ve
ikinci olarak morfolojik farklılıklar vasıtasıyla R. solani’ ye dayanıklılık açısından
taranmıştır. Yapılan analizler sonucu Russet Burbank (tuza toleranslı, R. solani’ ye
dayanıklı), Desiree (tuza toleranslı, R. solani’ ye hassas), Granola (tuza hassas R. solani’ ye
dayanıklı) ve Lady Claire (hem tuza hem de R. solani’ ye hassas) çeşitleri seçilmiş ve üçüncü
aşama olan; kontrol, NaCl, R. solani, R. solani+NaCl interaksiyonu uygulamalarına maruz
bırakılmıştır. Çeşitlerin olgunlaşma dönemlerinin ayrı ayrı stres faktörlerine ve
kombinasyonuna tolerans göstermeleri üzerinde etkili olduğu düşünülmektedir.
Karbonhidratların varlığı ve dağılımının R. solani+NaCl stresine toleransta etkili olduğu
belirlenmiştir. Yapılan analizlerde gerek enzim aktivitesi gerekse gen ifade düzeyi sonuçları
değerlendirildiğinde özellikle yumru büyümesi ve olgunlaşması dönemlerinde invertazların
daha yüksek seviyelerde ifade edildiği tespit edilmiştir. Bitki savunma ve yanıt
mekanizmalarındaki rolleri de düşünüldüğünde NaCl, R. solani, R. solani+ NaCl streslerinde
invertazların sukroz sintaza kıyasla daha önemli bir rol oynadıkları belirlenmiştir. Bununla
birlikte yaklaşık 73, 45 ve 35 kDa moleküler ağırlığa sahip üç dehidrin proteinin NaCl, R.
solani, R. solani+ NaCl streslerine toleransta etkili olduğu saptanmıştır
Climate change alone affects yields and nutritional values of plants, as well as
causing the spread of pathogens by negatively affecting the habitats of plant pests and
pathogens. For this reason, plants must evolve to live where they are exposed to different
stress factors at the same time. In this study, it was aimed to determine the changes in the
physiological characteristics and sucrose metabolism of the potato varieties that show
different responses to salt stress, Rhizoctonia solani and the combination of these two stress
factors at the molecular and gene expression level. For this purpose, in the experiment
planned as three stages, 19 commercial potato cultivars were screened fistly for salinity
tolerance by physiological and morphological differences along with yield and yield
components, and secondly for resistance to R. solani by morphological differences.
According to the results of the screenings, Russet Burbank (NaCl tolerant, R. solani
resistant), Desiree (NaCl tolerant, R. solani sensitive), Granola (NaCl sensitive, R. solani
resistant) and Lady Claire (susceptible to both) cultivars were selected and subjected to the
third stage; control, NaCl, R. solani, R. solani+NaCl interaction applications. Results
showed that the maturity periods of the varieties influence tolerance of individual stress
factors and combination. The presence and distribution of carbohydrates was determined to
be effective in tolerance to R. solani+NaCl stress. When the results of both enzyme activity
and gene expression were evaluated, it was determined that invertases were expressed at
higher levels, especially during the tuber initiation and bulking stages. Considering their
roles in plant defense and response mechanisms, invertases are thought to play a more
important role in NaCl, R. solani, R. solani + NaCl stresses compared to sucrose synthase.
