Bu çalısmada, Kargı Tulum Peynir’inden izole edilen laktik asit bakterileri
(LAB) tanımlanarak, bunların peynir fermentasyonunda starter kültür olma özellikleri
arastırılmıstır.
Yedi Kargı tulum peynirinden toplam 96 LAB izolatı elde edilmistir. Đzolatlar,
API CH50 ve API 20Strep identifikasyon sistemi ile tanımlanmıstır. Buna göre; 22
izolat Lactobacillus plantarum, 11 izolat Lb. brevis, 9 izolat Lb. paracasei subsp.
paracasei, 3 izolat Lb. pentosus, 3 izolat Lb. fermentum, 1 izolat Lb. curvatus subsp.
curvatus, 1 izolat Lb. rhamnosus, 9 izolat Enterococcus durans, 5 izolat E. faecium, 3
izolat Streptococcus thermophilus, 1 izolat St. mitis, 1 izolat St. equinus olarak
tanımlanmıstır. 8 izolat E. faecium/durans, 1 izolat E. faecium/gallinarum, 1 izolat E.
faecium/faecalis, 1 izolat E. gallinarum/casseliflavus ve 1 izolat St.
acidominimus/pluranimalium genus düzeyinde tanımlanabilmistir. Aerococcus viridans
(1), Granulicatella adiacens (1), Gemella morbillorum (7) gibi LAB olmayan izolatlar
bulunmustur. Đzolatlardan altı tanesi tanımlanamamıstır.
Lb. plantarum (5, 7), E. durans (103), E. faecium (71) suslarının, yüksek asit
üretimleri (6 saatte) ve asidik pH değerlerine dirençli olmaları nedeniyle, tulum
peynirinin üretiminde starter kültür olarak kullanımı uygundur. Buna ek olarak Lb.
pentosus (15), Lb. plantarum (3, 8, 9), E. faecium (112), Lb. brevis (46) gibi suslar
güçlü proteolitik aktiviteye sahip olmaları, asidik pH’a dirençli olmaları ve dekstran
ve/veya diasetil üretmeleri nedeniyle bu listeye eklenebilir.
Lb. brevis (37, 44), Lb. paracasei subsp. paracasei (43) and E. faecium (71);
insanlardaki biyolojik engellere (% 1-3 safra tuzu, 1-3 aralığındaki pH değerleri, oksijen
varlığı/yokluğu) dayanıklılıkları, bazı antibiyotiklere dirençli olmaları, laktaz, H2O2,
diasetil ve/veya bakteriyosin benzeri maddeler üretmeleri nedeniyle, probiyotik kültür
olarak en fazla ümit vaad eden suslardır.
The present work was aimed at identifying strains of Lactic Acid Bacteria
(LAB) from Kargı Tulum Cheese, with properties suitable for use as starter cultures in
cheese fermentation.
A total of 96 LAB isolates were obtained from seven samples of Kargı tulum
cheese. The isolates were identified by API 50CH and API 20Strep identification
systems and shown to be, 22 strains of Lactobacillus plantarum, 11 strains of Lb.
brevis, 9 strains of Lb. paracasei subsp. paracasei, 3 strains of Lb. pentosus, 3 strains of
Lb. fermentum, 1 strain of Lb.curvatus subsp. curvatus, 1 strain of Lb. rhamnosus, 9
strains of Enterococcus durans, 5 strains of E. faecium, 3 strains of Streptococcus
thermophilus, 1 strain of St. mitis, 1 strain of St. equinus, 8 strains of E. faecium/durans,
1 strain of E. faecium/gallinarum, 1 strain of E. faecium/faecalis, 1 strain of E.
gallinarum/casseliflavus and 1 strain of St. acidominimus/pluranimalium. There were
non lactic acid bacteria identified such as Aerococcus viridans (1), Granulicatella
adiacens (1), Gemella morbillorum (7). Six of the isolates could not be identified.
Strains of 5, 7 (Lb. plantarum), 103 (E. durans) ve 71 (E. faecium) could be
suitable as starter culture for tulum cheese fermentation because of its strong acid
production (in 6 h) and high acid tolerance. Additionally, Lb. pentosus (15), Lb.
plantarum (3, 8, 9), E. faecium (112), Lb. brevis (46) can be added to this list because
of its high proteolytic activity, acid tolerance, dextran and/or diacetyl production.
Lb. brevis (37, 44), Lb. paracasei subsp. paracasei (43) and E. faecium (71) can
be selected as the most promising strains as probiotic culture because of their surviving
in the human biological barriers (1-3% bile salt, pH values between 1-3, presence or
absence of oxygen), resistance to some antibiotics in general use and can produce,
lactase, H2O2, diacetyl and/or some bacteriocin-like substances.
