Kanser, dünyada ölümcül hastalıklardan biridir ve glial tümörler santral sinir sistemi
tümörlerinin en büyük grubudur. Kanser tedavisinde kemoterapi, radyoterapi ve cerrahi
yöntemler uygulanır. Ancak, çoğu tedavi konakçının normal hücrelerine de zarar verir.
Bu yüzden, kanser tedavisinde doğal ürünlerin kullanılmasına izin verilmiştir.
Mantarlar, yüzyıllar boyunca yiyecek olarak tüketildiği gibi birçok hastalığın tedavisi
için ilaç olarak kullanılmışlardır. Çalışmamızda altı adet yenilebilir mantar türünden
(Coprinus comatus, Fistulina hepatica, Lentinus strigosus, Laetiporus sulphureus,
Polyporus squamosus ve Lenzites betulina) elde edilen özütlerin sıçan glioma hücre
dizisi (C6) üzerindeki etkileri araştırılmıştır. C6 hücreleri 37 ºC’de, % 5 CO2 bulunan in
vitro ortamda çoğaltıldı. Hücreler daha sonra 96 kuyucuklu kültür kaplarına ekilerek
0.4, 1, 2, 4 ve 6 μg/mL ekzopolisakkarit dozları ile inkübe edildi. 24 ve 48 saat sonunda
ekzopolisakkaritlerin etkisi 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium
bromide, (MTT) yöntemi ile belirlendi. İstatistiksel analizler tek yönlü varyans
analizinin (ANOVA) ardından Tukey’in çoklu karşılaştırma yöntemi ile yapıldı. Elde
edilen sonuçlara göre, 0.4 μg/mL dozunda etki gözlenmemiştir. Ancak, 1, 2, 4 ve 6
μg/mL dozlarının glioma hücrelerini % 35’den % 80’ e kadar öldürdüğü bulunmuştur.
Sonuç olarak, ekzopolisakkaritlerin glioma hücrelerini zamana ve doza bağlı olarak
öldürdüğü tespit edilmiştir.
Cancer is one of the major wordwide fatal diseases and glial tumors are the
largest group of central nervous system tumors. Chemotherapic, radiotherapic and
surgical methods exist for the treatment of cancer in modern medicine however, most
cancer therapeutic methods severely affect the host normal cells. Hence, the use of
natural products now has been contemplated of exceptional value in the control of
cancer. Mushrooms have been used for many centuries, not just as a food, but also to
treat many illnesses. In this study the effects of exopolysaccharides from edible
mushroom species (Coprinus comatus, Fistulina hepatica, Lentinus strigosus,
Laetiporus sulphureus, Polyporus squamosus and Lenzites betulina) were investigated
on the rat glioma cell line (C6). C6 cells were cultured in a humidified atmosphere of 5
% CO2 at 37 0C in flasks. The C6 cells were then seeded in 96 well plates and exposed
to 0.4, 1, 2, 4 ve 6 μg/mL fungal exopolysaccharides doses. After 24 or 48 hours, the
action of each exopolysaccharides dose was measured by using 3-(4,5-dimethylthiazol-2-
yl)-2,5-diphenyltetrazolium bromide, (MTT) assay. All statistical analyses were
performed by one-way analysis of variance (ANOVA) and followed by Tukey’s
multiple comparison tests. According to our results 0.4 μg/mL was not effective, but 1,
2, 4 and 6 μg/mL doses killed glioma cells about % 35 to % 80. As a result, these
mushroom exopolysaccharides were time and dose dependently toxic to glioma cells.
