Amaç: Bu çalısmada, Türk meme kanserli kadın hastalarda (n=98) ve kontrol grubunda
(n=56), FGFR2 genindeki rs2981582 ve TNRC9 genindeki rs3803662
polimorfizmlerini taramak ve bu polimorfizmlerin meme kanseri riski ile iliskisini
belirlemek amaçlanmıstır.
Metaryal ve Metot: Çalısmada, Bursa Ali Osman Sönmez Onkoloji hastanesinde tanı
konulan 98 meme kanseri hastasından ve ailesinde meme kanseri hikayesi olmayan 56
kontrol bireyinden alınan periferik kan örnekleri kullanıldı. FGFR2 genindeki
rs2981582 ve TNRC9 genindeki rs3803662 SNP’lerinin belirlenmesi için PZR-RFLP
yöntemi kullanıldı.
Bulgular: Yapılan analiz sonucunda, 98 hastanın FGFR2 genotip dağılımı CC
(wildtype), CT (heterozigot) ve TT (polimorfik) genotipleri için sırasıyla, %39.8
(n=39), %49 (n=48), %11.2 (n=11) olarak bulundu. TNRC9 genotip dağılımı ise CC
(wildtype), CT (heterozigot) ve TT (polimorfik) genotipleri için sırasıyla, %41.8
(n=41), %49 (n=48), %9.1 (n=9) olarak bulundu. Hastalar ve kontrol grubu arasında her
iki polimorfizm için genotipik farklılık gözlenmedi (p>0.05). Östrojen reseptör
durumuna göre hastalar karsılastırıldığında her iki polimorfizmde T allel sıklığı dağılımı
açısından ER pozitif hastalar ER negatif hastalara göre istatistiksel olarak anlamlı
düzeyde yüksek bulundu (P<0.05). Meme kanseri hastalarının evre ve grad özellikleri
arasında yapılan karsılastırmalarda da genotip dağılımları arasında anlamlı bir farklılık
bulunmadı.
Sonuç: Bu çalısma Türk meme kanseri hastalarında, FGFR2 ve TNRC9 gen
polimorfizmlerinin arastırıldığı ilk çalısmadır. Meme kanseri riski ile FGFR2 ve
TNRC9 polimorfizmleri arasındaki bağlantının daha büyük populasyonlarda ve daha
ayrıntılı çalısılması meme kanserine yatkınlığı olan bireylerin belirlenmesine önemli
katkılar sağlayacaktır.
Purpose: The aim of the study was to determine the rs2981582 polymorphism exists in
FGFR2 gene and rs3803662 polymorphism exists in TNRC9 gene for Turkish breast
cancer patients (n=98) and healthy controls (n=56) and to investigate the relation
between breast cancer risk.
Materials and Methods: In this study, the peripheral blood samples were used which
were obtained from both 98 breast cancer patients and healthy controls whom were
diagnosed in Bursa Ali Osman Sönmez Oncology Hospital. The PZR-RFLP method
was used to determine the rs2981582 polymorphism exists in FGFR2 gene and
rs3803662 polymorphism exists in TNRC9 gene.
Results: According to the analysis that have been made, the genotypic distribution for
FGFR2 of the 98 patients for CC (wildtype), CT (heterozygote) and TT (polymorphic)
genotypes were found to be respectively, % 39,8 (n=39),%49 (n=48) and %11,2 (n=11).
The genotypic distribution for TNRC9 of the 98 patients for CC (wildtype), CT
(heterozygote) and TT (polymorphic) genotypes were found to be respectively, % 41,8
(n=41), %49 (n=48) and %9,1 (n=9).No genotypical difference was observed for both
polymorphisms between patients and healthy controls (p>0,05). As the patients were
compared according to ER status, for both polymorphisms, statistically ER positive
patients were more significant than ER negative patients according to T allel frequency
(P<0.05). No signicant difference was observed between the genotypical distrubution
during the comparation of stage and grade characteristics of breast cancer patients.
Conclusion: This is the first study investigating the FGFR2 and TNRC9 gene
polymorphism in Turkish breast cancer patients. Studying the relation of the breast
cancer risk and FGFR2 and TNRC9 polymorphisms on bigger populations in a more
detailed way can greatly help to determine the people who have breast cancer
susceptibility.
