Bu çalısmada, degisik dozlarda etanol ve aspirinin birlikte kullanımının rat beyin
sinaptozomları üzerine olan in vitro etkisi ve betainin olası koruyucu yolu arastırıldı. Bu
amaçla yirmi bir adet Spraque Dawley cinsi erkek albino rat dekapite edilerek
öldürüldü. Frontal korteksleri (ön beyin) alındıktan sonra her ön beyin dört parçaya
bölündü. Bu doku örneklerinden on dört grup olusturuldu ve her bir grup altı adet ön
beyin parçası içeriyordu (n=6). Sinaptozomal fraksiyonlar ön beyin parçalarının
homojenizasyon ve santrifüj asamaları sonrasında elde edildi. Etanol (50, 100, 200mM),
aspirin (100μg/ml) ve betainin (0,5, 1mM) varlıgında inkübe edilen sinaptozamal
fraksiyonlarda siyalik asit (SA) ve nitrik oksit (NO) düzeyleri ile adenozin deaminaz
(ADA) aktiviteleri ölçüldü.
Farklı akut dozlarda in vitro olarak uygulanan etanol doza baglı olarak rat beyin
sinaptozomları üzerinde nörotoksik etki göstermektedir. 100mM ve 200mM dozlarda
uygulanan etanol, SA parametresinde gösterildigi gibi sinaptozomal zarlarda hasar
yaratabilmektedir. 200mM dozda uygulanan etanol, NO ve ADA parametrelerinin
gösterdigi gibi fonksiyonel metabolik yollarda degisimlere sebep olmaktadır. Buna baglı
olarak zarda protein yapıdaki reseptör ve enzimlerin fonksiyonları bozulabilmektedir.
100μg/ml dozda aspirin, SA ve NO düzeylerinde herhangi bir degisiklige neden
olmamıstır. Etanol ile aspirin (100μg/ml) beraber uygulandıgı zaman olusan hasar
etanol dozuna baglı olarak SA ve NO düzeylerinde toksik etkiyi artırabilmektedir.
Betainin ise doza baglı olarak aspirin ile etanolün birlikte kullanımıyla artan
sitotoksiteyi azaltabilecegini düsünüyoruz.
This study deals with the in vitro effects of different doses of ethanol when it
is used with aspirin on rat brain synaptosomes as well as the possible protective
pathway of betaine. With this aim, twenty one male albino rats of Spraque Dawley
type are killed by decapitation. After the frontal cortexes of the rats are taken out,
each of them is divided into four pieces. Fourteen groups of which including 6
frontal cortex pieces each are generated from these tissue samples (n=6). The
synaptosomal fractions are prepared by the homogenization of the frontal cortex
pieces and centrifugation. Sialic acid (SA), nitric oxide (NO) levels and adenosine
deaminase (ADA) activities of synaptosomal fractions incubated with ethanol (50,
100, 200mM), aspirin (100μg/ml) and betaine (0.5, 1mM) are recorded.
In vitro administration of different doses of ethanol have neurotoxic effect
on rat brain synaptosomes. 100mM and 200mM ethanol, as it is shown in the SA
parameter, might cause damage at synaptosomal membrane and also
administration of 200mM ethanol may result in functional changes at metabolic
pathways, which is shown by the NO and ADA parameters. Due to these changes,
the structure and as a result of this, the function of the receptors and enzymes may
be disrupted. The administration of 100μg/ml aspirin does not cause any change at
SA and NO levels. When ethanol and aspirin (100μg/ml) are applied together, the
toxicity at SA and NO levels might increase according to the doses of ethanol. On
the other hand we think that betaine may decrease the cytotoxicity that is caused
by the usage of ethanol and aspirin, together.
