Vazektomi, spermatogenezin durmasına ve seminifer tübüllerin
dejenerasyona sebep olduğundan tüm dünyada yaygın olarak kullanılan bir korunma
yöntemidir. Bu çalısmadaki amaç, eriskin Wistar Albino sıçanlarda testis yapılarını
olusturan germ ve destek hücrelerinin sayısını ve diğer morfometrik özelliklerini ortaya
koyarak deneysel unilateral vazektominin etkilerini açıklamaktadır.
Erkek 8 haftalık Wistar Albino sıçanlara 1, 12 ve 24 hafta ve bunların
kontrolleri olmak üzere üç grup halinde unilateral vazektomi yapılmıstır. Hayvanların
testisleri ısık mikroskobik ve morfometrik incelenmeler için perfüzyon ile alındı.
Tarafsız ve etkin stereolojik araçlar ve stereolojik yöntemler kullanıldı. Optik disektör,
destek hücreleri ve germ hücrelerinin toplam sayısını hesaplamada kullanıldı. Cavalieri
yöntemiyle testisin toplam hacim ve hacim oranları hesaplandı.
Seminifer epitelin bütün hücrelerinin sayı ve yoğunlukları 24. haftalık
vazektomi grubunda önemli derecede azalmıstır. Vazektominin ilk etkilenen
spermatidlerin sayı ve yoğunlukları tüm vazektomi gruplarında azalalmıstır. 1 ve
12.haftalarda destek hücreleri ve spermatogonyumların toplam sayılarında değisiklik
olmamıstır. Testisin tüm hacmi bütün vazektomi gruplarında azalmıs olmasına rağmen
interstisyum hacmi değismedi. 12 ve 24. haftalarda Đnterstisyumun hacim oranının
artmasına rağmen adlüminal ve bazal kompartımanların hacim oranları azaldı.
Lümenlerin genislemesi interstisyumun artmasına pareleldir. Ayrıca ilk vasektomi
grubunun baslangıcından itibaren testislerdeki bazal membranlar kalınlastı. Đnterstisyel
alanda mononükleer hücre infiltrasyonu ve damarlanma artısı da 12. ve 24. haftadan
itibaren artmıstır.
Bulgularımız ısığında, 1 ve 12. haftalarda etkili olan telafi mekanizmasının 24
haftada yetersiz kaldığı ileri sürülebilir. Biz 12. haftadan sonra interstisyum ve bazal
membrandaki artısın, testisdeki hücrelerin etkilesimini bozduğunu düsünmekteyiz.
Vasectomy is a contraceptive method widely used around the globe, causing
degeneration of seminiferous tubules, and interruption of spermatogenesis. This study
was performed to determine the effects of experimental unilateral vasectomy on germ
cell and supporting cell counts, along with other morphometric characteristics of
testicular structures in adult Wistar Albino rats.
Normal male Wistar Albino rats, aged 8 weeks, were subjected to unilateral
vasectomy in three groups of 1, 12, 24 weeks and their control. Testes of animals were
perfused for light microscopic and morphometric investigations. Unbiased and efficient
stereological tools and stereological methods were used. An Optical dissector was used
for counting the total number of supporting and germ cells. Total volumes and volume
fraction of testes were estimated with the Cavalieri principle.
Number and numerical density of all cells of the seminifer epithelium
decreased significantly in 24-week vasectomy group. Number and numerical density of
spermatid is decreased in all of the vasectomy groups which. Total number of
supporting cells and spermatogonium did not change 1 and 12 weeks. Although total
volume of testes decreased in all of the vasectomy groups, volume of interstitial tissue
did not change. Relative volume of adluminal and basal compartments decreased in 12
and 24 week groups, in spite of the increased ratio of interstitial tissue volume. The
expansion of the lumen has been parallel to the increase in interstitial tissue.
Furthermore, the basal membranes were thickened in testes starting with the first
vasectomy group. Increased mononuclear cell infiltration and vascularization were also
observed in the interstitial space following 12 and 24 weeks.
Implications: Our data suggest that the compensation mechanism that are
effective in 1 and 12 weeks fails in 24 weeks. We believe that the increase in the
interstitial tissue and basement membrane damage cell interaction in testes starting
after 12 weeks.
