Alkol metabolizmasının %90‟ının karaciğerde gerçekleĢmesinden dolayı uzun süre alkol kullanımının karaciğer hasarına yol açtığı bilinmektedir. Alkol, karaciğerdeki doğal antioksidanların seviyelerini azaltarak hasar meydana getirir ve bu hasar antioksidanların kullanımı ile kısmen de olsa önlenebilir.
Karaciğer doku hasarına karĢı günümüzde doğal kaynaklı ilaçların kullanımı gün geçtikçe yaygınlaĢmaktadır. Makrofungusların ürettikleri ve büyüme ortamına salgıladıkları ekzopolisakkaritlerin (EPS) tıbbi açıdan birçok biyolojik aktiviteye sahip oldukları bilinmektedir. Bu nedenle araĢtırmamızda Coprinus comatus türünden elde edilen EPS‟nin, 3 farklı dozu uygulandı. Wistar albino cinsi sıçanlar kullanılarak; Grup I (Kontrol), Grup II (Alkol grubu), Grup III (Alkol + 50 mg/kg EPS), Grup IV (Alkol + 100 mg/kg EPS) ve Grup V (Alkol + 150 mg/kg EPS) oluĢturuldu. ÇalıĢma sonunda biyokimyasal, sitolojik ve histolojik etkiler değerlendirildi. Alanin transaminaz (ALT), laktat dehidrogenaz (LDH) enzimlerinin serum seviyeleri ile karaciğer dokularında kaspaz 3 ve sitokrom c oksidaz aktiviteleri ile mitokondri iç ve dıĢ zar bütünlükleri belirlendi. Elde ettiğimiz veriler sonucunda, Grup I‟e göre, Grup II‟de serum ALT seviyelerinde bir artıĢ gözlenirken LDH seviyesinde bir azalma tespit edildi. Karaciğer kaspaz 3 aktiviteleri gruplar arasında farklı bulunmadı. Alkol grubu olan Grup II‟ye ait sitokrom c oksidaz aktivitesinin, kontrol grubuna göre belirgin biçimde azaldığı tespit edildi. Grup II‟de mitokondrial transmembran potansiyeli düĢük bulundu. EPS uygulanan gruplarda ise iç zar yükünde, Grup II‟ye göre istatistiksel olarak anlamlı artmalar gözlendi (p<0.05). Alkol grubu karaciğer hepatositlerinde dejenerasyon, sinüzoidal alanlarda geniĢlemeler gözlendi. 50 mg/kg uygulanan EPS dozunda bu bulgular minimal düzeyde belirlendi. Elektron mikroskobu değerlendirmelerinde Grup II‟de mitokondride bozulmalar, yağlanma ve endoplazmik retikulumda ĢiĢme tespit edildi. 50 mg/kg uygulanan EPS dozunda ise kısmi olarak korunduğu görüldü. ÇalıĢmamız sonucunda Coprinus comatus’un 50 mg/kg dozunun alkole bağlı karaciğer hasarını tedavi edici etkisi olduğunu söylemek mümkündür.
It is known that, since %90 of the alcohol metabolism takes place in liver, long-term alcohol usage causes liver damage. Alcohol causes damage by reducing the level of natural antioxidants and this damage may be prevented by the usage of antioxidants. Against liver tissue damage, usage of natural origin drugs is becoming widespread contemporarily. It is known that, exopolisaccaharides (EPS), which macrofungus produce and secrete to the growing media, have many biological avtivity medically. For this reason, in our study, three different dosages of EPS, which were obtained from Coprinus comatus, were applied. Via by using albino type rats, Group I (Control), Group II (Alcohol Group), Group II (Alcohol + 50 mg/kg EPS), Group IV (Alcohol + 100 mg/kg EPS) and Group V (alcohol + 150 mg/kg EPS) were formed. At the end of the study, biochemical, cytological and histological effects were evaluated. Alanine transaminase (ALT), lactate dehydrogenase (LHD) enzymes‟ serum levels, and caspases 3 activities, cytochrome c oxidase and mitochondria‟s inner and outer membrane integrity in liver tissues were determined. As the result of data we obtained, while an increase was observed in bottom serum levels of Group II with regard to Group I, a decrease was determined in LHD level. Liver caspases 3 activities weren‟t found to be different between groups. It was determined that, cytochrome c oxidase activity of Group II, which is an alcohol group, decreased siginificantly. In Group II, mitochondrial transmembrane potantiel was found to be low. As for groups that were applied EPS, in inner membrane charge, statistically significant increase ( p<0.05) was observed with regard to Group II. In alcohol group liver hepatocytes, degeneration was observed, and in sinusoidal areas expansion was observed. In 50 mg/kg applied EPS dosage, these findings were determined in minimum level. In Electron Microscopical evaluations, in Group II, corruption and lipoidosis in mitochondria and expansion in reticulum was determined. As for 50 mg/kg applied EPS, it was seen that, it was conserved partially. As the result of our study, it is possible to say that, 50 mg/kg dosage of Coprinus comatus has therapeutic effect for alcohol dependent liver damage.
