Tezimizde
azoospermik ve oligozoospermik erkek hastalarda hem kromozom anomalilerinin
hem de Y kromozom mikrodelesyonlarının sıklığını ve tiplerini belirlemek; saptanan
genetik anomaliler ile infertil erkeklerdeki klinik veriler arasındaki ilişkiyi
incelemektir. Genetik anomali saptanan azoospermik ve oligozoospermik
erkeklerde, genetik anomali varlığının intrasitoplazmik sperm injeksiyonu (ICSI)
sonrası fertilizasyon üzerine etkisini araştırmaktır. Çalışmamız 97 infertil ve 10
kontrol fertil erkek üzerinde yürütüldü. Y kromozom mikrodelesyonu saptanan bazı
hastaların babalarında da aynı durumun varlığı araştırılmıştır. Çalışmaya dahil edilen
97 infertil erkek arasında azoospermik olan 73 olgunun 2’inde (~%3) hastada Y
kromozom mikrodelesyonu saptanmıştır. Azoospermik olan 73 hastadan 13’ünde ve
oligozoospermik 24 hastanın ise 3’ünde toplam 16 infertil hastada kromozom
anomalisi saptanmıştır. Y kromozom mikrodelesyonu ve kromozom anomalisi sıklığı
açısından araştırıldığında infertil erkek popülasyonda toplam genetik anomali sıklığı
olarak belirlenmiştir. Genetik anomalisi olan vakaların ICSI sonrası gebelik sonuçları
ile genetik anomalisi olmayan vakaların ICSI sonrası gebelik sonuçları arasında
istatistiksel olarak anlamlı bir fark bulunmamıştır. Fertil olduğu bilinen ve kontrol
grubunu oluşturan 10 kişide ise ne Y kromozomu mikrodelesyonu ne de kromozom
anomalisi saptanmamıştır.
In our thesis, is to determine both the frequency and types of
chromosome abnormalities and Y chromosome microdeletion in male patients with
azoospermic and oligozoospermic and to detect the relationship between clinical data
of infertile males determined genetic abnormalities. Genetic abnormalities detected
in men with azoospermic and oligozoospermic, is to investigate impact of the
presence of genetic abnormalities on outcome of fertilization through after ICSI. In
our thesis is performed 97 infertile patients and 10 control fertile male. The same
situation was also investigated in fathers of patients with Y chromosome
microdeletions. The study of 97 infertile male, 73 were azoospermic and 24 were
oligozoospermic. Totally 2 patients detected Y chromosome microdeletions; 2 out of
73 azoospermic patients had Y chromosome microdeletions. Totally 16 patients
detected chromosome abnormalities; 13 out of 73 in patients with azoospermic and 3
out of 24 in patients with oligozoospermic detected chromosomal abnormalities. We
investigate the incidence of Y chromosome microdeletion and chromosomal
abnormalities and the incidence of genetic abnormalities in the infertil male
population. Among patients who had pregnancy after ICSI, there was no correlation
found between patients those has genetic abnormality and the patients those hasn’t.
We detected neither Y chromosome deletion nor chromosomal abnormalities in the
fertile control group of 10 person.
