Bu çalışmamızda bir monoterpen olan izoforon molekülünün Alternaria
alternata fungal kültürü ile biyotransformasyonu ve oluşan metabolitlerin
antimikrobiyal etkisi incelendi.
İzoforon’un Alternaria alternata ile biyotransformasyon sonucu iki farklı mono
hidroksillenmiş metabolit ürettiği ince tabaka kromatografisi yardımıyla tespit edildi.
Metabolitler kolon kromatografisi yardımıyla saflaştırıldı ve yapıları spektroskopik
yöntemler (GC–MS, ¹H–NMR, ¹³C–NMR, HSQC ve HMBC) kullanılarak (S)-4-
hidroksi–3,5,5-trimetil-2-siklohegzen-1-on ve 3-hidroksimetil-5,5-dimetil-2-
siklohegzen-1-on olarak tanımlandı.
Elde edilen metabolitlerin disk difüzyon metodu kullanılarak antimikrobiyal
incelemeleri sonucu Acinetobacter baumanii, Escherichia coli, MRSA (metisiline
dirençli Staphylococcus aureus) ve Bacillus subtilis mikroorganizmalarına karşı
antibakteriyal etki gösterdiği tespit edildi. Minimum inhibisyon konsantrasyonu
değerleri sırasıyla M1 metaboliti için 9,4; 37,4; 37,5 ve 18,7 mg/ml, M2 metaboliti için
10,5; 21,0; 21,0 ve 21,0 mg/ml olarak belirlendi.
The aim was to investigate microbial transformation of the monoterpene,
isophorone, using the pathogenic fungi Alternaria alternata and determine the antimicrobial
activities of the metabolites.
Biotransformation of isophorone with Alternaria alternata yielded two mono
hydroxylated derivatives as detected by thin layer chromatography. The metabolites
were purified by silica gel column chromatography and their structures were identified
as (S)-4-hydroxy–3,5,5-trimethyl-2-cyclohekzen-1-one and 3-hydroxymethyl-5,5-
dimethyl-2-cyclohekzen-1-one on the basis of GC-MS, ¹H–NMR, ¹³C–NMR, HSQC
and HMBC spectra.
Antimicrobial tests using disc diffusion methods indicated that M1 and M2
showed good antibacterial activities against Acinetobacter baumanii, Escherichia coli,
MRSA (methycilline resistent Staphylococcus aureus) and Bacillus subtilis. The
Minimum Inhibition Concentrations were calculated as 9,4; 37,4; 37,5 ve 18,7 mg/ml
for M1 and 10,5; 21,0; 21,0 ve 21,0 mg/ml for M2, respectively.
