Çalışmamızda, Hypericum perforatum, Hypericum aviculariifolium, Hypericum adenotrichum ve Hypericum sechmenii ekstraktlarının, Lipopolisakkarit (LPS) ile inflamasyon oluşturulan sıçanlar üzerine olan etkileri araştırıldı. Bu amaçla, 4-5 aylık, 70 adet Wistar albino ırkı dişi sıçan 10 gruba ayrıldı. İnflamasyonun oluşturulması için; inflamasyon, inflamasyon+H. perforatum, inflamasyon+H. adenotrichum, inflamasyon+ H. aviculariifolium ve inflamasyon+H. sechmenii gruplarındaki sıçanlara 1 mg/kg dozunda LPS serum fizyolojik içerisinde çözülerek verildikten sonra, ilgili ekstraktlar 7 gün boyunca 150 mg/kg olarak gavaj yoluyla verildi. Kontrol gruplarına ise serum fizyolojik, H. perforatum, H. adenotrichum, H. aviculariifolium ve H. sechmenii verildi. Sıçanların karaciğer, akciğer ve böbrek F-18-florodeoksiglukoz (FDG) pozitron emisyon tomografi (PET) taramaları yapılıp, 18FDG tutulumları sud maks olarak hesaplandı. Görüntülemenin ardından hayvanlar, ketamin/ksilazin anestezisi altında kalp kanı, karaciğer, böbrek dokuları alındı. Eritrosit hemolizatlarında malondialdehit (MDA), süperoksit dismutaz (SOD), katalaz (KAT), nükleer faktör kappa β (NF-kβ), tümör nekroz faktör (TNF-α), İnterlökin-1β (IL-1) ve İnterlökin-6 (IL-6), karaciğer ve böbrek doku homojenatlarında MDA, SOD, KAT, NF-kβ ve TNF-α seviyeleri ELISA yöntemiyle belirlendi.
Yapılan çalışma sonucunda, LPS uygulanan inflamasyon gruplarında, MDA, NF-κβ, TNF-α, IL-1β ve IL-6 seviyelerinde artma, SOD ve KAT seviyelerinde azalma tespit edildi. H. perforatum, H. adenotrichum, H. aviculariifolium ve H. sechmenii ekstraktları inflamasyon gruplarında, karaciğer MDA, SOD, KAT, NF-κβ ve TNF-α, böbrek MDA, NF-κβ ve TNF-α, hemolizat MDA, SOD, TNF-α ve IL-6 seviyelerini kontrol grubuna yaklaştırdığı belirlendi. H. sechmenii’nin, bakılan bütün biyokimyasal parametreleri kontrol grubuna yaklaştırdığı belirlenirken, H. aviculariifolium’un, hemolizat KAT, NF-κβ, IL-1β, H. adenotrichum’un, böbrek KAT, hemolizat NF-κβ, H. perforatum’ un, böbrek KAT seviyesi üzerine etkisi görülmedi.
Akciğerde, H. sechmenii’nin 18FDG tutulumunu kontrol seviyesine yaklaştırdığı belirlendi. Akciğer, karaciğer ve böbrek FDG-PET analizlerinde, H. perforatum, H.
v
In our study, the effects of Hypericum perforatum, Hypericum aviculariifolium, Hypericum adenotrichum and Hypericum sechmenii extracts on Lipopolysaccharides (LPS) induced inflammation in rats was investigated. For this purpose, 4-5 months old, 70 Wistar albino rats were divided into 10 groups. To induce inflammation; 1 mg / kg LPS was administrated to inflammation, inflammation+H. perforatum, inflammation+H. adenotrichum, inflammation+H. aviculariifolium and inflammation+H. sechmenii groups, intraperitoneally, which was followed by treatment of 150 mg/ kg related extracts for 7 days by gavage to LPS induced groups, whereas saline, H. perforatum, H. adenotrichum, H. aviculariifolium and H. sechmenii was administrated as control groups. Fluoro-18-deoxyglucose positron emission tomography (18FDG-PET) scans of rats’ liver, lung and kidney were performed and 18FDG uptake was calculated as sud max. Following imaging, hemolysate, liver and kidney tissues were obtained under ketamine / xylazine anesthesia. In erythrocyte hemolysates, the levels of malondialdehyde (MDA), superoxide dismutase (SOD), catalase (KAT), nuclear factor kappa-β (NF-kβ), tumor necrosis factor-α (TNF-α), interleukin-1 β (IL-1β) and interleukin-6 (IL-6), in liver and kidney homogenates the levels of MDA, SOD, KAT, NF-kβ and TNF-α were determined by ELISA method.
As a result of the study, an increase in the levels of MDA, NF-κβ, TNF-α, IL-1β and IL-6, and a decrease in the levels of SOD and KAT were detected in LPS induced inflammation groups. Administration of H. perforatum, H. adenotrichum, H. aviculariifolium and H. sechmenii extracts recovered liver MDA, SOD, KAT, NF-κβ, TNF-α, kidney MDA, NF-κβ, TNF-α, and hemolysate MDA, SOD, TNF-α, IL-6 to the control levels. while H. sechmenii recovered all biochemical parameters to the control group, there was no effect of H. aviculariifolium on hemolysate KAT, NF-κβ, IL-1β, H. adenotrichum on kidney KAT, hemolysate NF-κβ levels, and H. perforatum on kidney KAT level.
In the lung, it was determined that H. sechmenii recovered completely lung18FDG uptake to the control level. 18FDP-PET analysis of lung, liver and kidney
vii
was indicated that H. perforatum, H. adenotrichum, H. aviculariifolium and H. sechmenii reduced inflammation.
In conclusion, it can be deducted that H. adenotrichum, H. aviculariifolium and H. sechmenii have similar effects compared to H. perforatum when the inflammatory and oxidative effects caused by LPS were evaluated by liver, kidney, hemolysate parameters and FDG-PET analysis
