Antioksidanlar, normal metabolizma sırasında üretilen bazı reaktif oksijen türlerinin vücuda verdiği yoğun zararı azaltabilen veya tamamen ortadan kaldırabilen maddelerdir. Genelde bitkisel kaynaklar doğal antioksidan olarak kullanılırken, son yıllarda ekstrem ortamlardan izole edilen metabolitlerinde bu amaçla kullanılabileceği çeşitli çalışmalarla gösterilmiştir. Ekstrem bir ortam olan tuzlu ortamlardan izole edilen mikrofunguslar, yeni antioksidan maddeler için doğal bir kaynak oluşturmaktadırlar.
Bu çalışmada, tuzlu ortamlardan izole edilen mikrofungusların antioksidan özelliklerinin belirlenebilmesi için, Folin-Ciocalteu kolorimetrik metodu kullanılarak toplam fenolik madde içeriği, DPPH (2,2-difenilpikrilhidrazil) üzerinden serbest radikal süpürücü etki ve antioksidan enzimlerden olan süperoksit dismutaz (SOD) ve katalaz (KAT) spesifik enzim aktivitesi araştırılmıştır. Serbest radikal süpürücü etki tayini için üç farklı konsantrasyon (9.6×10-4, 1.8×10-3 ve 3.6×10-3 mg/mL) kullanılmıştır. Ayrıca serbest radikal süpürücü etki ince tabaka kromotografisi ile değerlendirilmiştir. Toplam fenolik madde miktar tayininde; 1 (%98), 9 (%82), 17 (%50), 5 (%45) ve 15 (33) kodlu izolatlar yüksek aktivite göstermiştir. Serbest radikal süpürücü etki açısından ise; 1 (%86), 2 (%69), 7 (%67), 8 (%78), 9 (%76), 13 (%79), 15 (%78) dir. Süperoksit dismutaz, spesifik enzim aktivitesi 20 adet fungustan 12 tanesinde (izolat numaraları sırasıyla; 1, 4, 5, 6, 7, 8, 9, 11, 12, 18, 19 ve 20) yüksek bulurken, spesifik katalaz aktivitesi 16 tanesin de (1, 4, 5, 6, 8, 9, 10, 11, 13, 14, 15, 16, 17, 18, 19 ve 20) yüksek bulunmuştur.
Sonuçlara göre, tuzlu ortamlardan izole edilen funguslar serbest radikal süpürücü aktivite ve spesifik enzim aktiviteleri açısından korelasyon göstermektedir.
Bu çalışma yeni doğal antioksidan maddelerin keşfi bakımından önem taşımaktadır.
Antioxidants, which are produced during normal metabolism, reactive oxygen species in some of the extensive damage to the body-killing substances that can reduce or completely. Generally used as vegetable sources of natural antioxidants, extreme environments in recent years, several studies have demonstrated that isolated metabolites can be used for his purpose. Microfungi isolated from the salty environments of extreme environments, constitue a natural resorce for new antioxidant substances.
In this study, the antioxidant properties of microfungi isolated from saline environments in order to determine the Total Phenolic content, DPPH (2,2difenilpikrilhidrazil) via free radical scavening effect and antioxidant enzymes superoxide dismutase and catalase, the enzyme specific activity was investigated. For the determination of free radical scavenging effect of three different concentrations (9.6×10-4, 1.8×10-3 and 3.6×10-3 mg/ml) were used. In addition, free radical scavenging effect was evaluted thin-layer chromatography. Quantification of total phenolic compounds; 1 (%98), 9 (%82), 17 (%50), 5 (%45) and 15 (33) coded isolates showed high activity. In terms of the free radical scavenging effect; 1 (%86), 2 (%69), 7 (%67), 8 (%78), 9 (%76), 13 (%79), 15 (%78) is. Superoxide dismutase, a specific enzyme activity in fungi out of 20 12 of them (numbers of isolates, respectively; 1, 4, 5, 6, 7, 8, 9, 11, 12, 18, 19 and 20) high, while the specific catalase activity and 16 (1, 4, 5, 6, 8, 9, 10, 11, 13, 14, 15, 16, 17, 18, 19 and 20) were higher.
According to the results, free radical scavenging activity of fungi isolated from salty environments, and had a correlation specific enzyme activities. This study is important for the discovery of new natural antioxidant substances.
