Bu tez çalışmasında sıçanlarda deneysel olarak oluşturulan karaciğer iskemi/reperfüzyon (İR) hasarına karşı, antioksidan özelliği bilinen gallik asitin olası koruyucu etkileri araştırıldı.
Çalışmada 28 adet 3-4 aylık Spraque-dawley cinsi dişi sıçanlar kullanıldı (n=7).
Bunlar rastgele seçimle 4 gruba ayrıldı. Grup I (Sham Grubu), Grup II (İR + Serum Fizyolojik), Grup III (İR + 50 mg/kg gallik asit), Grup IV (İR + 100 mg/kg gallik asit) olarak belirlendi. Grup I ve II’e serum fizyolojik (1 ml/kg), Grup III ve IV’e sırasıyla 50 mg/kg, 100 mg/kg dozlarındaki gallik asit, (serum fizyolojik içinde çözdürülerek 1 ml/kg) 1 hafta boyunca günde 1 kere oral yolla verildi. Grup I’e laparotomi yapıldı ve hepatik arter, portal ven ve safra kanalları çevre dokulardan temizlendikten sonra başka hiçbir işlem uygulanmadan kapatıldı. Grup II, III ve IV için iskemi süresi 45 dakika, reperfüzyon süresi ise 60 dakika olarak belirlendi. Deney sonunda kan serumunda Alanin aminotransferaz (ALT), Aspartat aminotransferaz (AST) aktivitelerine ve karaciğer örneklerinde Katalaz (CAT) ve Glutatyon Peroksidaz (Gpx) enzim aktiviteleri ölçüldü. Doku kesitleri Hematoksilen&Eozin ile boyandı ve ışık mikroskobunda
incelendi.
Çalışma sonunda İ/R hasarının, Grup I ile Grup II ‘nin ALT ve AST değerleri karşılaştırıldığında Grup II’de yükseldiği ve bunun sonucunda İ/R hasarının oluşturulduğu gözlendi. Gallik asit uygulaması sonucunda CAT ve Gpx aktivitelerinin kontrol değerine yaklaşarak hasarı önlediği görüldü. Buna göre 50 mg/kg gallik asitin, 100 mg/kg gallik asite göre daha etkili olduğu düşünüldü. Elde edilen histolojik
bulgular da bu sonuçları destekler nitelikteydi.
Çalışmanın sonucu karaciğer İR hasarına gallik asitin koruyucu etkili olduğunu gösterdi.
In this thesis study, possible protective effects of gallic acid, which is known to be antioxidant, were investigated against experimentally induced liver ischemia/reperfusion (IR) injury in rats.
In the study, three to four month, Spraque-dawley type 28 female rats were used (n=7).
They were separated to 4 groups randomly. Groups were determined as follows: Group I (Sham
Group), Group II (IR + normal saline), Group III (IR + 50 mg/kg gallic acid), Group IV (IR +
100 mg/kg gallic acid). Groups I and II were inoculated normal saline (1 ml/kg), Group II and IV were inoculated 50 mg/kg and 100 mg/kg of gallic acid respectively (via solving in normal saline, 1 ml/kg) through oral routine once a day for one week. Laparotomy was performed to Group I, hepatic artery, portal vein and bile ducts were cleaned from peripheral tissues and then closed without any further operations. For Group II, II and IV, ischemia and reperfusion durations were determined to be 45 mins and 60 mins respectively. At the end of the experiment, alanine aminotransferase (ALT), aspartate aminotransferase (AST) activities in blood, Catalase (CAT) and glutathione peroxidase (Gpx) enzyme activities in liver samples were measured. Histological sections were stained by Hematoxylen & Eosin and investigated by light microscope.
At the end of the study, when Group I and Group II’s ALT and AST values of (I/R) injury were compared, it was determined that there was an increase in Group II and as a result of this, I/R injury was formed. It was observed that, injury recovered by CAT and GPx, which became less active by gallic acid, and has approached to control value. 50 mg/kg gallic acid was thought to be more effective when compared to 100 mg/kg gallic acid. Histological findings were supporting these results too.
Result of the study showed that gallic acid has protective effect liver I/R injury.
