I/R hasarı oluşturulan sıçan karaciğerinde,
rotenonun koruyucu etkisini immünohistokimyasal yöntemlerle değerlendirmeyi
amaçladık. Bu deneysel çalışmada 30 adet Spraque-Dawley cinsi albino sıçan
kullanıldı. Sıçanlar Kontrol Grubu, I/R Grubu ve I/R+Rotenon Grubu olacak şekilde
3 eşit gruba ayrıldı. Kontrol grubundaki sıçanlara 8 saat açlık sonrası laparatomi
uygulanarak hepatik arterler, portal ven ve safra yolu eksplore edilip, 90 dakika
sonra kan örneği ve karaciğerden doku örneği alındı.I/R grubundaki sıçanlara; 45
dakika iskemi gerçekleştirildi ve reperfüzyon’dan 45 dakika sonra kan ve karaciğer
örneği alındı.. I/R+Rotenon grubundaki sıçanlara iskemiden 2 saat önce rotenon
intraperitoneal olarak verildi. 45 dakika reperfüzyon sonrasında kan ve
karaciğerden doku örneği alındı.Daha sonra tüm sıçanlar sakrifiye edildi. Serumda
aspartat aminotransferaz (AST), alanin aminotransferaz (ALT), laktat dehidrogenaz
(LDH) ve malonildialdehit (MDA) seviyeleri, dokuda ise MDA ve TNF-α düzeyleri
ile biyokimyasal, ışık mikroskobunda Hematoksilen&Eozin boyama ile
histopatolojik, TUNEL boyama yöntemiyle de apoptozis üzerine değerlendirmeler
yapıldı. Serum ALT, AST, LDH ve MDA düzeyleri, doku MDA ve TNF-α
düzeylerinin hepsi I/R grubunda yükselirken rotenon ile tedavi edilen grupta
(I/R+Rotenon) anlamlı olarak gerilemiştir (p< 0.05). Konjesyon ve polimorf nüveli
lökositler gibi histopatolojik bulgular da I/R+Rotenon grubunda gerilemiştir.
TUNEL boyamasında sayılan apoptotik hücrelerin sayısı da I/R grubunda artarken,
I/R+ Rotenon grubunda anlamlı olarak gerilemiştir (p<0.05). Yaptığımız çalışma
göstermiştir ki; rotenon , antioksidan ve antiapopitotik etkilerinden dolayı karaciğer
I/R hasarını önlemek amacıyla kullanılabilir
Purposes; to evaluate the protective effects of rotenon on liver following
I/R injury, by immunehistochemical methods . Thirty Spraque-Dawley rats were
used in this experimental study. Animals were divided into 3 groups including the
Control Group, the I/R Group, and the I/R+ Rotenon Group. In the Control group,
the arteries , portal veins and main biliary ducts were explored and after 90 minutes,
blood and liver tissue samples were collected. In the I/R group, after the arteries ,
portal veins and main biliary ducts were clamped for 45 minutes, blood and liver
tissues samples were collected. In the I/R+Rotenon group, rotenon was administered
intraperitoneally before 2 hours of ischemia and blood and liver tissues samples were
collected 45 minutes later. All rats were sacrificed at the end of the experiment. The
aspartat aminotransferase (AST), alanin aminotransferase (ALT), lactate
dehydrogenase (LDH) and malonedialdehyde (MDA) levels of blood samples and
the TNF-α and malonedialdehyde (MDA) levels of tissue samples biochemically and
tissue samples histopathologically under light microscopy with hematoxylin and
eosin stain, apoptosis TUNEL assay were studied. The blood ALT, AST, LDH and
MDA levels and the tissue MDA and TNF-α levels were increased in the I/R group
but in the rotenon treatment group (I/R+Rotenon) were decreased significantly (p<
0.05). Histopathological findings such as congestion and polymorphonuclear
leukocytes were observed less in the I/R+Rotenon group compared to the I/R group.
In the TUNEL assay, the number of the apoptotic cells were increased in the I/R
group however decreased in the I/R+Rotenon group significantly(p<0.05). In our
study, we showed that rotenon with antioxidant and antiapoptotic properties may be
used in surgery clinics for liver I/R injury.
