Erişkin erkek sıçanlarda lityum karbonat ile oluşturulan testis hasarı üzerine E
vitamininin etkisi.
Lityum 19. yüzyıldan bu yana tıpta kullanılan alkali bir iyondur. Lityum karbonat
bipolar bozuklukların akut atak tedavisinde ve uzun dönem profilaksisinde ilk seçenek
olan ajanlardan biridir ve terapotik alanı çok dardır. Fakat uzun dönemde terapotik
dozlarda kullanımı bile üreme sistemini de kapsayan çeşitli organlarda ve sistemlerde
bir çok yan etkiye neden olmaktadır.
E vitamini, biyolojik membranlarda serbest radikal hasarını engelleyen güçlü bir
antioksidandır. Bu çalışmada lityum karbonat ile oluşturulan sıçan testis hasarına karşı
E vitamininin etkilerinin ortaya konulması amacıyla 28 adet Spraque-Dawley türü sıçan
kullanıldı. Sıçanlar her grupta 7 erişkin erkek sıçan olacak şekilde kontrol, 100 mg/kg
lityum karbonat, 100 mg/kg E vitamini, 100 mg/kg lityum karbonat+100 mg/kg E
vitamini grubu olmak üzere 4 gruba ayrıldı. 20 günlük deney sonunda sıçanların vücut
ve testis ağırlıkları ölçüldü ve ardından karşılaştırmalar yapıldı. Sol testisler doku takip
işlemi için Bouin çözeltisi içerisine, sağ testisler ise %10‘luk nötral formalin içerisine
alındı ve rutin histolojik işlemlerden sonra bloklandı. Elde edilen parafin bloklardan 3
μm kalınlığında seri kesitler alındı ve kesitler Hematoksilin+Eozin ve Periyodik Asit-
Schiff+ Hematoksilin ile boyanarak mikroskobik incelemeleri yapıldı.
Sonuç olarak lityum karbonatın testis ve vücut ağırlığını azalttığı, testiste
seminifer tübüllerde ve tübül hücrelerinde hasara yol açtığı, ayrıca tübül lümenindeki
kuyruklu sperm sayısını azalttığı gözlendi. Lityum karbonat ile birlikte verilen E
vitaminin ise testiste lityum karbonatın oluşturduğu hasarı azalttığı gözlendi.
Effect of vitamin E on testicular damage induced by lithium carbonate in adult
male rats.
Lithium is an alkaline ion being used since 19th century. Lithium is one of the
first choice agents for long-term prophylaxis and the treatment of acute episodes of
bipolar disorders and its therapeutic index is extremely narrow. However, ın long
periods of use, even the therapeutic doses, can cause several side effects in various
organs and systems including the reproductive system.
Vitamin E is a powerful antioxidant preventing the propagation of free radical
damage in biological membranes. In this study, 28 Sprague- Dawley rats were used
determine the effects of vitamin E on lithium carbonate induced testicular damage in
rats. in the present study. Rats were divided into 4 groups as control, 100 mg/kg lithium
carbonate, 100 mg/kg vitamin E, 100 mg/kg lithium carbonate +100 mg/kg vitamin E,
with 7 adult male rats in each group. At the end of administration period, body and
testes weights were measured and compared to each other. Left testes were put into
Bouin solution, right testes were put into 10% neutral formalin for tissue processing and
blocked after the routine histological procedures. Serial sections at 3μm thickness were
obtained from those paraffin blocks and microscopical examinations were performed on
the testicular sections after staining by Hematoxyline + Eosine and Periodic Acid-
Schiff+ Hematoxyline.
Results of the present study indicated that, lithium carbonate causes reduction in
the body and testes weights, damages the seminiferous tubules and tubular cells and
decreases the number of tailed sperms. However, it was observed that administration of
vitamin E together with lithium carbonate might lead to reduction in damage the caused
by lithium carbonate in testes.
