Erişkin erkek sıçanlarda metotreksat kaynaklı testis hasarında kurkuminin etkisi

Abstract

Metotreksat, kanser tedavisinde sıklıkla kullanılan kemoterapotik bir maddedir. Ayrıca testis üzerinde de toksik bir etki göstermektedir. Hücre ve dokular, meydana gelen bu toksik etkilerden kurkumin gibi antioksidan sistemlerle korunabilirler. Bu çalışmada, metotreksat ile oluşturulan sıçan testis hasarına karşı kurkuminin etkilerinin ortaya konulması amacıyla 28 adet Spraque-Dawley türü sıçan kullanıldı. Sıçanlar her grupta 7 erişkin erkek sıçan olacak şekilde kontrol, 20 mg/kg metotreksat, 200 mg/kg kurkumin, 20 mg/kg metotreksat+200 mg/kg kurkumin verilen grup olmak üzere 4 gruba ayrıldı. Deney sonunda sıçanların vücut ve testis ağırlıkları ölçüldü ve karşılaştırmalar yapıldı. Sol testisler doku takip işlemi için Bouin çözeltisi içerisine, sağ testisler ise %10’ luk formaldehit içerisine alındı ve rutin histolojik işlemlerden sonra bloklandı. Elde edilen parafin bloklardan 3 μm kalınlığında seri kesitler alındı ve testislerden alınan kesitler Hematoksilin + Eozin ve Periyodik Asit-Schiff+Hematoksilin ile boyanarak mikroskobik incelemeleri yapıldı. Çalışmanın sonucunda elde ettiğimiz bulgulara göre, metotreksatın vücut ağırlığını azalttığı, testiste seminifer tübüllerde ve hücrelerde hasara yol açarak spermatogenezi etkilediği gözlendi. Metotreksat ile birlikte verilen kurkuminin ise testiste oluşan bu hasarı azalttığı gözlendi.

Methotrexate is a frequently used chemotherapeutic agent, it also has toxic effect on testes. Cells and tissues may be protected from these toxic effects by antioxidant substances like curcumin. In this study, 28 Sprague - Dawley rats were used to determine the effects of curcumin on metotrexate induced testicular damage in rats. Rats were divided into 4 groups as control, 20 mg/kg methotrexate, 200 mg/kg curcumin, 20 mg/kg methotrexate + 200 mg/kg curcumin, including 7 adult male rats in each group. At the end of administration period, body and testes weights were measured and compared to each other. Left testes were put into Bouin solution, right testes were put into 10% formaldehyde for tissue processing and blocked after the routine histological procedures. Serial sections with 3μm thickness were obtained from those paraffin blocks and microscopical examinations were performed on sections of testes after stained by Hematoxyline + Eosine and Periodic Acid- Schiff + Hematoxyline. Results showed that methotrexate causes the loss of body weight in rats. Methotrexate also affects the spermatogenesis by damaging the seminiferous tubule and cells. On the other hand, it was observed that administration of curcumin together with methotrexate might reduce this damage in rat testicle.
