Sıçanlarda akut karaciğer hasarına,
leptinin koruyucu etkisini immünohistokimyasal yöntemlerle apoptozis üzerinden
değerlendirmek. Yöntemler: Bu deneysel çalışmada 18 adet Spraque-Dawley cinsi
albino sıçan kullanıldı. Sıçanlar Kontrol Grubu, CCl4 Grubu ve Leptin + CCl4 Grubu
olacak şekilde 3 eşit gruba ayrıldı. Kontrol grubuna 0.8 ml/kg zeytinyağı
intraperitoneal (i.p.) yolla uygulandı, CCl4 ve CCl4+leptin gruplarına ise 0.8 ml/kg
CCl4 (1.1 oranında zeytiyağı ile çözülmüş) i.p. yolla uygulandı. Uygulamadan 6 saat
sonra CCl4+leptin grubuna 10 µg/kg Leptin i.p. yolla uygulandı. Son uygulamadan
24 saat sonra bütün sıçanlar sakrifiye edildi. Serumda aspartat aminotransferaz
(AST), alanin aminotransferaz (ALT), alkalen fosfataz (ALP) ve tümör nekrozis
faktör alfa (TNF-α) seviyeleri, dokuda ise malonildialdehit (MDA) ve TNF-α
düzeyleri ile biyokimyasal, ışık mikroskobunda Hematoksilen&Eozin boyama ile
histopatolojik, TUNEL boyama yöntemiyle de apoptozis üzerine değerlendirmeler
yapıldı. Sonuçlar: Serum ALT, AST, ALP ve TNF-α düzeyleri, doku MDA ve TNF α düzeylerinin hepsi CCl4 grubunda yükselirken leptin ile tedavi edilen grupta
(CCl4+leptin) anlamlı olarak gerilemiştir (p< 0.05). Konjesyon, polimorf nüveli
lökositler ve balon dejenerasyonu gibi histopatolojik bulgular da CCl4+leptin
grubunda gerilemiştir. TUNEL boyamasında sayılan apoptotik hücrelerin sayısı da
CCl4 grubunda artarken CCl4+ leptin grubunda anlamlı olarak gerilemiştir (p<0.05).
Karar: Yaptığımız çalışma göstermiştir ki CCl4 ile oluşturulan karaciğer hasarında;
leptin tedavisi karaciğeri oldukça başarılı bir şekilde korumuştur. Bu nedenle aşırı
apoptozis sonucu ortaya çıkabilecek istenmeyen bazı tablolarda leptinden tedavi
edici bir ilaç olarak faydalanılabilir.
The aim of this study is to investigate the protective effect of leptin in
rats on CCl4 -induced acute liver damage using immunohistochemical methods for
apoptosis and biochemical parameters. Methods: In this experimental study, 18
albino rats from Spraque-Dawley genus were divided into three equal groups as;
control, CCl4 and CCl4+Leptin. 0.8 ml/kg olive oil was administered
intraperitoneally (i.p.) to the control group, 0.8 ml/kg CCl4 (1:1 dissolved in olive
oil) was administered i.p to the CCl4 and CCl4+leptin groups. Six hours after the
treatment 10 µg/kg Leptin was administered i.p. to the CCl4+leptin group. Twenty four hours after administrating CCl4 all of the groups were sacrificed. Biochemical
assessments were performed using serum aspartate aminotransferase (AST), alanine
aminotransferase (ALT), alkaline phosphatase (ALP), tumor necrosis factor alfa
(TNF-α) levels and tissue malondialdehyde (MDA) and TNF-α levels.
Histopathological assessments were performed using Hematoxilin&Eosin staining in
light microscope and apoptosis assesment using TUNEL. Results: Serum ALT,
AST, ALP and TNF-α levels, tissue MDA and TNF-α levels were all increased in
CCl4 group, but were significantly decreased in group treated with leptin
(CCl4+Leptin) (p<0.01). Histopathological comparison of the groups showed a
decrease in congestion, polymorphonuclear leukocytes and hepatocyte ballooning in
CCL4+leptin group. TUNEL-positive cell counts in liver were significantly increased
in CCl4 group, but decreased in CCl4+Leptin group (p< 0,05). Conclusion: The
results of the present study indicate that leptin treatment substantially prevents CCl4-
induced apoptosis in the liver. Thus, leptin may serve as a drug for treating many
clinical conditions that arise from inappropriate apoptosis.
