Bu tez çalısmasında; sıçanlarda, deneysel karaciğer iskemi/reperfüzyon (Đ/R)
hasarı sonucu olusan oksidatif strese karsı sumak (Rhus coriaria)’ın olası koruyucu
etkileri arastırıldı.
35 adet disi Spraque Dawley sıçan rasgele seçilerek; Grup I (Đ/R), Grup II
(Kontrol), Grup III (Đ/R + 50mg.kg-1 sumak), Grup IV (Đ/R + 100mg.kg-1 sumak) ve
Grup V (Đ/R + 200mg.kg-1 sumak) olusturuldu. Ksilazin (10 mg.kg-1) ve ketamin (70
mg.kg-1) anestezisi altında Grup II dısındaki tüm gruplara total karaciğer iskemisi
yapıldı. Tedavi uygulaması iskemiden 30 dk önce tek doz intraperitoneal olarak yapıldı.
Ardından bu gruplara 45 dk iskemi / 60 dk reperfüzyon uygulandı. Deney sonunda tüm
gruplardan, kan örnekleri ve karaciğer dokuları alındı. Sonuçlar istatistiksel olarak
değerlendirildi. Kontrol grubuna göre Đ/R grubunda serum ALT ve AST değerlerinin ve
karaciğer dokusuna ait CAT aktivitelerinin arttığı; tedavi gruplarında doza bağlı olarak
bu değerlerde iyilesme tespit edildi. Histopatolojik analizlerde ise Đ/R grubunda
karaciğer dokusunda hepatositlerde dejenerasyon, kanama alanlarında, yoğun nekroz,
sinusoidlerde genisleme, nüklear infiltrasyon ve hücresel vakuolizasyon gözlenmis
olup, 200 mg.kg-1 uygulamasına ait karaciğer dokusu incelendiğinde nekroz ve
vakuolizasyona rastlanmayıp, kanama da anlamlı düsüs gözlendi. Sonuç olarak bu
grupta hücre bütünlüğü korunmus olup, histopatolojik olarak kontrole yakın değerler
görüldü.
Çalısma, intraperitoneal olarak uygulanan sumak ekstresinin 200 mg.kg-1
dozunun karaciğer Đ/R hasarında koruyucu etkisi olduğunu gösterdi.
In this thesis study; possible protective effects of sumac (Rhus coriaria) was
investigated against oxidative stress which occurred as a result of induced liver
ischemia/reperfusion (I/R) injury in rats.
By randomly selecting 35 female Spraque Dawley rats; Group I (I/R), Group II
(Control), Group III (I/R + 50 mg.kg-1 sumac), Group IV (I/R + 100 mg.kg-1 sumac) and
Group V (I/R + 200 mg.kg-1 sumac) were formed. Except Group II, liver ischemia was
performed to all groups, under Xylazine (10 mg.kg-1) and ketamine (70 mg.kg-1)
anaesthesia. Treatment application was performed 30 min before the ischemia as a
single dose intraperitoneally. After that, 45 min of ischemia / 60 min of reperfusion was
applied to these groups. Blood samples and liver tissues were taken from all groups at
the end of the experiment. Results were evaluated statistically. It was determined that,
in I/R group, serum ALT and AST values, and CAT activities of liver tissue increased
with respect to control Group; in treatment group, a dose-dependent improvement was
determined for these values. In I/R group, degeneration of hepatocytes, congestion
areas, dense necrosis, expansion of sinusoids, nuclear infiltration and cellular
vacuolisation was observed in histopathological analysis of liver tissues. When liver
tissue belonging to 200 mg.kg-1 application was examined, necrosis and vacuolisation
wasn’t observed and a significant decrease in congestion areas was determined. As a
result, in this group cell integrity was protected and values close to control were
observed histopathologically.
Study showed that, intraperitoneally applied 200 mg.kg-1 dosage of sumac
extract has protective effect on liver I/R injury.
