Parkinson
Hastalığı, Alzheimer hastalığından sonra en sık görülen nörodejeneratif hastalıktır.
Son yıllarda genetik formlarının ve patojenik mutasyonların tanımlanması ile
Parkinson Hastalığının etiyopatogenezine ilişkin bilgiler artmıştır. Parkinson
hastalığında tanımlanan ilk gen SNCA genidir ve ilk nokta mutasyonlar da bu gende
tanımlanmıştır. LRRK2 geni G2019S mutasyonu da Parkinson Hastalığında en sık
gözlenen nokta mutasyondur. Çalışmamıza sporadik Parkinson Hastalığı tanısı
almış 83 olgu dahil edilmiştir. Parkinson Hastalığından sorumlu olduğu bilinen
SNCA genindeki A30P ve LRRK2 genindeki G2019S mutasyonlarının Eskişehir
bölgesinde tanı alan olgulardaki sıklığının belirlenmesi, tanısal amaçlı
kullanılabilirliği ve ayrıca elde edilen veriler ile moleküler etiyolojisi belirlenmiş
olan olgulara hastalığı ve mutasyonu ile ilgili genetik danışmanın verilmesi
amaçlanmıştır. Sekans analizi ile A30P mutasyonunun bulunduğu SNCA geni
ekzon 2 ve G2019S mutasyonunun bulunduğu LRRK2 geni ekzon 41 genomik
bölgeleri incelenmiş ve hastalarda herhangi bir nokta mutasyon ya da polimorfizm
saptanmamıştır. Çalışmamızda Eskişehir bölgesinde sporadik Parkinson Hastalığı
tanısı alan olgularda A30P ve G2019S mutasyonlarının sık görülmediği sonucuna
ulaşılmıştır.
Parkinson’s disease is the second most common
neurodegenerative disease after Alzheimer disease. Resently understanding genetic
forms and pathogenic mutations has been providing growing knowledges about
etiopathogenesis of Parkison’s disease. SNCA was the first gene with mutations
causing Parkinson’s disease. LRRK’ gene G2019S mutation is the most common
mutation causing Parkinson’s disease. In our study we investigate 83 patients with
sporadic Parkison’s disease. Aims of our study are to identify the frequency of the
SNCA A30P and LRRK2 G2019S mutations in Parkinson’s disease patients from
the Eskisehir region of Turkey, diagnostic utility of these mutations and to confer
genetic counselling to the mutation carier patients. SNCA exon 2 and LRRK2 exon
41 was investigated with direct squencing method. Neither any point mutation or
polymorphism was detected in the SNCA exon 2 or LRRK2 exon 41 from the
patients. Our findings suggest that the SNCA A30P and LRRK2 G2019S mutations
are not frequent in Turkish patients with Parkinson’s disease.
