Klorofenoller soğutucu, yangın geciktirici, boya çözucu, pestisit, herbisit ve kereste koruyucusu olarak cesitli alanlarda kullanilan kimyasal maddedir. Yüksek toksisiteleri, kanser oluşturabilmeleri ve dogada kolay bozunmamalari nedeni ile önemli kirleticiler olarak bilinirler. Klorofenollerin herhangi bir yolla organizmaya alinmasi ksenobiyotik (zenobiyotik) etki yaratir. Bazı mikroorganizmalar klorofenolleri biyolojik olarak bozundurup kloru ayristirip, organizmadan uzaklastirabilirler.Bu çalışmada, Trametes versicolor (ATCC, 200801) microorganizmasi kullanilarak, buğday kepeği (3 g/100 ml) ilaveli patates dekstrozlu broth ortamında, lakkaz enzimi üretilmiştir. Yüksek aktiviteli lakkaz enzimi kullanılarak, pH, substrat konsantrasyonu, enzim miktarı, inkübasyon süresi ve ortam sıcaklığı parametrelerinin 4-klorofenolden klor uzaklaştırılması işlemini uzerine olan etkisi incelenmiştir. Belirlenen optimum koşullarda 4- klorofenolün toksisitesindeki değişim Microtox cihazı ile takip edilmiştir. Ayrıca klor uzaklaştırıldıktan sonra 4-klorofenolün kimyasal yapısındaki değişiklikler FTIR analizleri ile belirlenmiştir.4-Klorofenolden klor uzaklaştırılması için uygun koşullar pH 4, başlangıç substrat konsantrasyonu 200 µM, süre 30 dk ve sıcaklık 40 oC ve enzim miktarı 4 ml olarak belirlenmiştir. Belirlenen optimum koşullarda oksijen tüketimi ve klor uzaklaştırılması arasındaki iliski araştırılmıştır. Mikrotoksisite sonuçlarına göre 5. dakikada yarım maksimal etkili konsantrasyon EC50 değerlerinde toksisitesinde bir artış görülmüştür. 4-klorofenolun bozundugunu belirten???..maddeleri bulunmustur.Çalışmamızdan elde edilen bulgular Trametes versicolor ATCC (200801)'den elde edilen lakkaz enziminin, 4-klorofenolden klor uzaklaştırılması isleminde etkin olarak kullanılabileceğini ortaya koymustur (ornegin: atik sularin aritilma toprak temızleme islemi gibi).
Chlorophenols, that are often used in the production of refrigeration cooling gas systems, fire retardants, paints, solvents, pesticides, herbicides and wood preservatives, are known to be an important class of xenobiotics. Because of their high toxicity, strong resistance to degradation and ability to build up in the body, chlorophenols are considered cause for cancer. Some microorganisms are able to degrate chlorophenols and produce dechlorination of chlorophenolics.In this study, laccase enzyme was produced through PDB culture including pellicle and Trametes versicolor (ATCC, 200801). The effect of proces parameters such as: pH , substrat concentration, enzyme concentration, reaction time and temperature on the degradation of 4-clorophenol was studied with laccase enzyme which had high activity for dechlorination of 4-chlorophenol. Correlation between oxygen consumption and dechlorination process was investigated in optimal media. Changes in toxicity versus the quantitiy of 4- chlorophenol were monitored using Microtox. Also, after the dechlorination of 4-chlorophenol, the changes in the chemical structure of 4-chlorophenol were determined with FTIR analysis.Optimal conditions for dechlorination of 4-clorophenol have been found to be: pH 4.0, substrat concentration 200 µM, reaction time 30 dk, temperature 40 °C and enzyme concentration 4 ml/L. During dechlorination studies, it was observed that the quantity of oxygen in the culture medium was decreased. Changes in the degree of toxicity of 4-chlorophenol was monitored using Microtox.
