Siklofosfamide bağlı sıçan testis hasarında E vitamininin rolünün histolojik olarak
incelenmesi.
Kemoterapötik bir madde olan siklofosfamid (CP), testis dokusu ve hücreleri
üzerinde toksik etki göstermektedir. Hücre ve dokular, meydana gelen bu toksik
etkilerden E vitamini (E vit) gibi antioksidan sistemlerle korunabilirler. Bu
çalışmamızda CP'nin sıçan testisleri üzerindeki toksik etkisi üzerine E vit'nin rolünü
araştırmayı amaçladık. Çalışmamızda, 32 adet Spraque-Dawley türü sıçan kullanıldı.
Sıçanlar her grupta 8 erişkin erkek sıçan olacak şekilde kontrol, 20 mg/kg
siklofosfamid, 20 mg/kg siklofosfamid + 200 mg/kg E vitamini ve 200 mg/kg E
vitamini verilen grup olmak üzere 4 gruba ayrıldı. Deney sonunda vücut ve testis
ağırlıkları ölçüldü ve karşılaştırmalar yapıldı. Sol testisler doku takip işlemi için Bouin
çözeltisi içerisine alındı ve rutin histolojik işlemlerden sonra bloklandı. Elde edilen
parafin bloklardan 3 µm kalınlığında seri kesitler alındı ve kesitler Hematoksilen,
Periyodik Asit-Schiff + Hematoksilen ve Toluidin mavisi ile boyanarak mikroskobik
incelemeleri yapıldı. Vücut ve testis ağırlıkları açısından gruplar arasında önemli fark
gözlendi. Mikroskobik incelemeler CP'nin testislerde hasara yol açtığını ve bu hasarın
CP+E vit verilen gruplarda azaldığını gösterdi. Elde edilen bulgulara göre, CP'ye bağlı
olarak testis dokusunda meydana gelen hasarın E vit verilmesiyle önlenebileceği
sonucuna varıldı.
Histological examination of the role of the vitamin E in cyclophosphamide induced testicular injury.
Cyclophosphamide (CP) is a chemotherapeutic agent but has also toxic effect on
testes. Testicular tissue and cells can be protected from toxic effects by antioxidant
systems, including vitamin E (vit E). Total 32 Sprague-Dawley rats were used in the
present study. Rats were divided into 4 groups as control, 20 mg/kg cyclophosphamide,
20 mg/kg cyclophosphamide + 200 mg/kg vitamin E and 200 mg/kg vitamin E, with 8
adult male rats in each. At the end of the treatment period, body weight and testes
weight were measured and comparisons were made. Left testes put into Bouin solution
for tissue tracking process and were blocked after the routine histological procedures.
Serial sections with a 3 µm thickness were obtained from that paraffin blocks, and
microscopical evaluations were made on sections stained with hematoxylene, periodic
acid-sciff + hematoxylene and toluidine blue. There was significant difference in body
weights and testes weights between the groups. Microscopical evalution revealed that
CP induced an injury in testes and that injury was less in the groups of animals treated
with CP + vitamin E. Results of the present study indicate that the testicular injury
induced by CP can be prevented by administration of vitamin E.
