anormal extrasellüler matriks materyalinin göz dokularında üretilmesi ve
birikmesi ile karakterize yaşa bağlı elastozis olarak tanımlanır.
Psödoeksfoliyasyon glokomu (PEG) ise sekonder açık açılı glokomun en sık
nedeni olup PES’da en sık görülen klinik tablodur. PES’nun patogenezi ve
psödoeksfoliyatif materyalin (PEM) biyokimyasal yapısı henüz tam olarak
bilinmemektedir. Son zamanlarda yapılan araştırmalarda, PES/PEG
olgularında 15. kromozomunun q24.1. lokusunda bulunan liziloksidaz-benzeri
protein 1 geninde (LOXL1) ki üç tek nükleotid polimorfizminin (SNP) varlığı
saptanmıştır. Çalışmamızda Türk toplumunda PES/PEG olgularında LOXL1
genindeki üç SNP’ini (rs2165241, rs3825942 ve rs1048661) araştırarak PES
ve PEG ile ilişkisini ortaya koymayı amaçladık. Bu nedenle çalışma
kapsamına alınan PES, PEG ve kontrol olgularının tam göz muayeneleri
yapıldıktan sonra kan örneklerinden DNA eldesi yapıldı. DNA örneklerinde
prob temelli bir genotipleme yöntemi olan RT-PCR ile polimorfizm varlığı
araştırıldı. Çalışma sonucunda PES/PEG olgularında, LOXL1 gen üç
SNP’nin kontrol grubu olgularından daha fazla olduğu saptandı. Ancak PES
ile PEG grubu arasında fark bulunamadı. Üç SNP’den PES/PEG ile en fazla
ilişkili olanı rs3825942 olup bu sonuç Avrupa ve Amerika’da yapılan
çalışmalar ile benzerlik göstermekteydi. Cinsiyete göre sonuçlar
değerlendirildiğinde sadece rs2165341 SNP’i erkeklerde 3 kat daha fazla
risk taşımaktaydı.

syndrome (PES) is defined as the age-related elastosis characterized by
production and accumulation of abnormal extracellular matrix materials in a
variety of ocular tissues. On the other hand, pseudoexfoliative glaucoma
(PEG) is the most common cause of secondary open-angle glaucoma and
the most common clinical picture seen in PES. PES pathogenesis and
biochemical structure of PEM is not known yet. In recent studies, in
PES/PEG cases, the presence of three single-nucleotide polymorphism
(SNPs) has been detected on lysyloxidase-like protein 1 (LOXL1) gene
located on chromosome 15 area q24.1. In our study, we aimed to investigate
the association of LOXL1 gene three SNPs (rs2165241, rs3825942 and
rs1048661) with PES/PEG cases in Turkish population and assess the
relationship with PES and PEG. For this purpose, after complete eye
examination of the PES, PEG and control cases included, DNA is extracted
from the blood samples and polymorphism is investigated by RT-PCR which
is a probe-based genotyping method. Based on the study results, LOXL1
gene three SNPs is more frequently seen in PES/PEG cases than it is seen
in control cases. However, no difference was found between PES and PEG
groups. Out of three SNP, rs3825942 was found to be more strongly
associated with PES/PEG than the other two and this was similar with the
studies conducted in Europe and America. When the results were evaluated
regarding the gender, the only significant finding was that, males carried
three times greater risk of having rs2165341 SNP.
