Tarımsal mücadelede gittikçe daha fazla tercih edilmeye başlayan biyokontrol
ajanlarının salgıladığı bazı enzimler, hastalık etmeni olan patojeni baskılayarak gelişimini
geriletmekte ve etkisini azaltmaktadır. Bu çalışmada önemli bir biyokontrol ajanı olan
Trichoderma spp.’nin, Rhizoctonia solani Kühn. fitopatojenine karşı gösterdiği enzimatik
tepki üzerinde durulmuştur. Materyal olarak R. solani ile T. viride (TVD1, TVD3), T. virens
(TVN1, TVN2, TVN3), T. harzianum (TH2, TH3, TH4) ve T. asperellum (TA1, TA2, TA3)
olmak üzere bazı Trichoderma türleri kullanılmıştır. Enzim üretim ortamında R. solani
bulunan ve bulunmayan (kontrol) iki grup Trichoderma spp.’nin selülaz enzim aktiviteleri
ayrı ayrı belirlenmiştir. Sonuçlar, R. solani bulunan örneklerin neredeyse tamamında, selülaz
enzim aktivitesinin kontrol grubuna oranla en az 2 katına çıktığını göstermiştir. Bu durum
Trichoderma spp.’nin, R. solani’yi baskılamak için biyokimyasal bir tepki oluşturduğunu
açıkça ortaya koymaktadır. Bu tepkinin morfolojik olarak da gösterilebilmesi adına karışık
ikili kültür tekniği kullanılmış; Trichoderma spp.’nin hızlı bir yayılım göstererek R.
solani’nin gelişim alanını sınırlandırdığı görülmüştür. Aktivite çalışmaları sırasında endo-ß 1,4-glukanaz enzim üretim ortamı için optimizasyon çalışmaları yapılmış; optimum sıcaklık
30°C, optimum pH 5,0 olarak belirlenmiş ve enzimin %1 (w/v) CMC’ye yüksek spesifite
gösterdiği görülmüştür. Sentezlenen fungal selülaz SDS-PAGE tekniği ile görüntülenmiştir.
Ayrıca pH, sıcaklık, farklı substratlar ve farklı substrat konsantrasyonları gibi parametrelerin
enzim aktivitesine etkisi de incelenmiştir. Elde edilen sonuçlar, Trichoderma spp.’nin
biyokimyasal savunma ve saldırı mekanizmasının, yeni ve çevre dostu ticari ürünlerin
geliştirilmesinde fikir verebileceğini belirtir niteliktedir
Some enzymes which released by biocontrol agents and preffered increasingly at
agricultural pest control, reduces the growth and the effect of the patogen which are disease
factor. In this research, enzymatic response of Trichoderma spp., an important biocontol
agent, against Rhizoctonia solani Kühn. phytopathogen was evaluated. Some Trichoderma
species as T. viride (TVD1, TVD3), T. virens (TVN1, TVN2, TVN3), T. harzianum (TH2,
TH3, TH4) and T. asperellum (TA1, TA2, TA3) and R. solani were used as material.
Cellulase enzyme activities of two Trichoderma spp. group that consist of R. solani and not
(control) in the enzyme production media were determined seperately. Results showed that
cellulase enzyme activities were nearly twice as much of the control group in almost all of
the samples that consist of R. solani. This result clearly reveals that the Trichoderma spp.
constitute a biochemical response to inhibit the R. solani. Dual culture technique was used
to represent this response morphologically and it was observed that Trichoderma spp.
showed a quick spread and limit the growth area of R. solani. During the activity researches,
optimization trials were carried out for the media of endo-ß-1,4-glucanase. Optimum
temperature and pH values were determined as 30°C and 5.0 respectively and high specifity
of the enzyme was observed to 1% (w/v) CMC. The synthesized fungal cellulase was
displayed via SDS-PAGE technique. Moreover, the effects of the parameters as pH,
temperature, different substrates and different concentrations of substrates on the enzyme
activity were also examined. The results indicated that biochemical defense and attack
mechanism of Trichoderma spp. may provide insight to develop new and eco-friendly
commercial products
