DNA, türlerin, tür gruplarının ve daha üst taksonların arasındaki evrimsel ilişkiyi
belirlemede kullanılan oldukça işlevsel bir araçtır. Bu çalışmada Palearktik’te geniş yayılış
gösteren Simulium cinsi ornatum tür grubunun Anadolu’da yayılış gösteren türlerinin
filogenileri, filocoğrafyaları ve taksonomik durumları COI, NADH2 ve ITS 1ve 2 gen
bölgeleri kullanılarak araştırılmıştır.
Türkiye’nin 19 farklı su havzası sınırları içindeki, 54 il ve 222 farklı lokaliteden
2014-2018 yılları arasında tür grubuna ait çok sayıda örnek elde edilmiştir. Bununla
birlikte Avrupa’daki 17 farkı lokaliteden toplanan bireyler de çalışmaya dâhil edilmiştir.
Toplam 384 bireyden total DNA eldesi gerçekleştirilmiştir. Polimeraz zincir reaksiyonu ve
Sanger sekanslama sonrasında her gen bölgesi için farklı sayıda veri setleri oluşturulmuş
ve veri setlerine; maksimum olasılık, maksimum parsimoni, Bayesian filogeni, haplotip ağı
analizleri, moleküler saat analizi ve genetik çeşitlilik analizleri uygulanarak tür grubunun
evrimsel ilişkilerinin ortaya çıkarılması amaçlanmıştır.
Çalışma sonucunda; morfotaksonomik olarak Simulium ornatum Meigen, 1818 ve
Simulium kiritshenkoi Rubtsov, 1940 olduğu tespit edilen türlerin genetik olarak iki ayrı tür
olmadığı tespit edilmiştir. Tür grubunun bir diğer türü olan Simulium intermedium
Roubaud, 1906 türü ülkemizden ilk kez kaydedilmiştir. Yapılan filogenetik analizler
sonucunda bu türün S. ornatum ve S. kiritshenkoi türlerinden farklılaşmakta olan kardeş bir
tür olarak değerlendirilebileceği ön görülmüştür. Ornatum tür grubu Anadolu türleri ile
ilgili faunistik verilerin revizyona ihtiyacı olduğu sonucuna varılmıştır. Geometrik
morfometri analizleri sonucunda morfolojik teşhislerde sık kullanılan pupa solunum
filamentlerinin, tür grubu için tür ayrımlarında işlevsiz olduğu görülmüştür. Tür grubu
içinde S. intermedium türünün, S. kiritshenkoi+S. ornatum kladı ile yaklaşık olarak 3,5
milyon yıl önce (Pleistosen-Pliyosen geçişi) son ortak atayı paylaştıkları tespit edilmiştir.
Genetik çeşitlilik analizleri sonucunda elde edilen verilerle tür grubunun Anadolu
populasyonları arasında hibritleşme oranının fazla olduğu sonucuna ulaşılmıştır
DNA is a highly functional tool for determining evolutionary relationship between
species, species groups and higher taxa. In this study, phylogeny, phylogeography and
taxonomy of the Anatolian S. ornatum species group, which is widely distributed in
Palearctic, were investigated using COI, NADH2 and ITS 1 and 2 regions.
A high number of specimens collected from 222 different localities representing 54
provinces in 19 freshwater basins in Turkey between 2014-2018. Besides, specimens from
17 different localities from Europe were included in the study. Total DNA was extracted
from 384 individuals. After polymerase chain reaction and Sanger sequencing, datasets
were generated for each region and; maximum likelyhood, maximum parsimony, Bayesian
phylogeny, haplotype network analysis, molecular clock analysis and genetic diversity
analysis were applied to the data sets with the aim of the assessment of the evolutionary
relationships of the species group.
The following conclusions were drawn for the species group;
morphotaxonomically, identfied as Simulium ornatum Meigen, 1818 and Simulium
kiritshenkoi Rubtsov, 1940 were found to be genetically not as two distinct species.
Simulium intermedium Roubaud, 1906, which is another species of the species group, was
recorded for the first time from our country. As a result of the phylogenetic analysis, it was
predicted that this species could be considered as a sibling species differentiating from S.
ornatum + S. kiritshenkoi clade. It is concluded that faunistic data of Ornatum species
group of Anatolia need revision. As a result of geometric morphometry analysis, pupae
respiratory filaments, which are frequently used in morphological diagnoses, were found to
be dysfunctional in species separation in this species group. Within the species group, S.
intermedium species, shared the last common ancestor with S. kiritshenkoi + S. ornatum
clade about 3.5 million years ago (Pleistocene-Pliocene transition). As a result of genetic
diversity analyzes, it is concluded that the hybridization rate of the species group among
Anatolian populations is very high
