Candida türleri önemli mukozal ve yayılıcı infeksiyonlara yol açabilen fırsatçı
mantarlardır. Son yıllarda bu infeksiyonların insidansında görülen artış ve yüksek mortalite
oranları, daha etkili antifungal tedavi stratejilerinin gerekliliğini ortaya koymaktadır. En
yaygın izole edilen tür C. albicans olmakla beraber, non-albicans türlerin yol açtığı
infeksiyonlar da sıklıkla rapor edilmektedir. Candida türleri morfolojik değişim ya da
biyofilm oluşumu gibi fenotipik adaptasyonları sağlayan Quorum Sensing (QS) molekülleri
salgılarlar. Bu sistemde, mikroorganizmalar ürettikleri sinyal molekülleri sayesinde
birbirleri ile haberleşebilmekte ve belirli bir yoğunluğa ulaştıklarında virülans faktörlerine
ait çeşitli genlerin ekspresyonlarını tetikleyebilmektedirler. Candida türlerindeki çoğunluk
algılama sistemi hakkında bilgiler oldukça azdır ve farnesol ökaryotlarda keşfedilen ilk QS
sinyal molekülüdür. İlginç bir şekilde farnesol, Candida türleri ile hem bir QS molekülü
olarak ve hem de ekzojen bir ajan olarak etkileşime girebilir. Farnesolün bu duyarlılığında
hücrenin gelişim süresi, boyutları, sıcaklığı ya da gelişim ortamı gibi faktörler etkili
olmaktadır. Çalışmamızda farnesolün hem planktonik ve hem de biyofilm formundaki
Candida türleri üzerine etkilerinin araştırılması hedeflenmiştir. Çalışmaya C. albicans’ın
yanısıra (1 standart ve 1 klinik izolat), 5 farklı klinik Candida izolatı dahil edilmiştir. Etken
maddenin planktonik Candida türleri üzerine etkisi broth mikrodilüsyon (MİK) ve geçirimli
elektron mikroskop (TEM) ile; biyofilm üzerine olan etkileri de kolorimetrik (XTT) ve
taramalı elektron mikroskop (SEM) ile araştırılmıştır. Sonuçlarımız, farnesolün Candida
türlerinin planktonik ve biyofilm formları üzerine yaklaşık 37,5 µM - 600 µM konsantrasyon
aralıklarındaki inhibe edici etkisini ortaya koymuştur. Farnesol daha düşük
konsantrasyonlarda ise biyofilmi artırıcı bir etki sergilemiştir. TEM verilerimiz; 37,5 µM
konsantrasyonda farnesol muamelesi sonucu hücrelerde membran ve duvar hasarları,
vakuolizasyon ya da granülasyonlar göstermiştir. SEM verilerinde ise farnesol muamelesi
sonucu pre ve post biyofilm uygulamalarında kontrol grubuna kıyasla biyofilm indirgenmesi
tespit edilmiştir. Farnesol antifungal ve antibiyofilm tedavi stratejilerinde etkili olarak
kullanılabilir. Ancak verilerimizin daha detaylı çalışmalarla desteklenmesi gerekmektedir
Candida species are opportunistic fungi that can cause significant mucosal and
invasive infections. The increase in the incidence of these infections in recent years and the
high mortality rates suggest the necessity of more effective antifungal treatment strategies.
Although the most common isolated species is C. albicans, infections caused by non albicans species are also frequently reported. Candida species secrete Quorum sensing (QS)
molecules that provide phenotypic adaptations such as morphological change or biofilm
formation. In this system, microorganisms can communicate with each other through the
signal molecules they produce and, when they reach a certain concentration, they can trigger
the expression of various genes of virulence factors. Information about the Quorum Sensing
system in Candida species is scarce and farnesol is the first QS signal molecule discovered
in eukaryotes. Interestingly, farnesol can interact with Candida species both as a QS
molecule and as an exogenous agent. Growth time and size of the cell, temperature or growth
environment are effective in this sensitivity of farnesol. The aim of this study was to
investigate the effects of farnesol on both the planktonic and biofilm forms of Candida
species. In addition to C. albicans (1 standard and 1 clinical isolate), 5 different clinical
isolates were included to the study. The effect of the active substance on planktonic Candida
was investigated by broth microdilution (MIC) and transmission electron microscope
(TEM); the effects on biofilm were also investigated by colorimetric (XTT) and scanning
electron microscope (SEM). Our results demonstrated the inhibitory effect of farnesol on the
planktonic and biofilm forms of Candida at concentrations ranging from about 37.5 µM to
600 µM. Farnesol showed a biofilm-enhancing effect at lower concentrations. Our TEM data
showed the membrane and wall damage, vacuolization or granulation in cells as a result of
farnesol treatment at a concentration of 37.5 µM. In SEM data, biofilm reduction was found
in pre and post biofilm applications compared to control group as a result of farnesol
treatment. Farnesol can be used effectively in antifungal and antibiotic treatment strategies,
but our data needs to be supported by more detailed studies
