Bu çalışmada benzimidazolyum bromür türevi olan Bileşik 1 ve Bileşik 2’nin 10, 25,
50, 75, 100, 150, 200, 250 ve 500 µM konsantrasyonlarının insan prostat kanseri hücreleri
(LNCaP) üzerindeki morfolojik etkisi incelenmiş ayrıca 3-(4,5-dimetiltiazol-2-il)-2,5
difeniltetrazolium bromid (MTT) yöntemi ile zamana ve konsantrasyon değişimine bağlı
sitotoksik etkisi araştırılmıştır. Pozitif kontrol olarak kullanılan 5-Florourasil (5-FU) ile
MTT sonuçlarına göre daha sitotoksik olduğu belirlenen Bileşik 1 konsantrasyonları 1:1
oranında kombine olarak LNCaP hücrelerine uygulanmış ve hücre çoğalması üzerindeki
antogonistik ve sinerjistik etkileri araştırılmıştır. Son olarak, ELISA (Enzyme Linked
Immunosorbent Assay) yöntemi kullanılarak Apoptotik DNA Fragmantasyonu
belirlenmiştir.
Bileşik 1’in 24 saatte 100 µM, 48 saatte ise 25 µM konsantrasyonundan itibaren
hücre morfolojisi üzerine etki etmeye başladığı gözlenmiştir. IC50 değerleri 24 ve 48 saat
için sırasıyla 183 ve 90 µM olarak hesaplanmıştır. Bileşik 2’nin 24 ve 48 saatte 50 µM
konsantrasyonundan itibaren hücre morfolojisi üzerine etki etmeye başladığı gözlenmiştir.
IC50 değerleri 24 ve 48 saat için sırasıyla 488 ve 239 µM olarak hesaplanmıştır. Pozitif
kontrol olarak kullanılan 5-FU’ın denenen konsantrasyonlarında ise 24 ve 48 saatte IC50
değeri bulunamamıştır. Kombine uygulamasında (Bileşik 1+5-FU), 24 saatte 200 µM, 48
saatte ise 100 µM konsantrasyonundan sonra antagonistik etkinin başladığı gözlenmiştir.
Bileşik 1’in 100 ve 250 µM konsantrasyonlarının LNCaP hücrelerindeki Apoptotik DNA
Fragmantasyonu araştırıldığında nekrotik etki görülmezken apoptotik etkinin
konsantrasyona bağlı olarak arttığı yani hücre ölümünün apoptozla gerçekleştiği
görülmüştür
In this study, the morphological effects of Compound 1 and Compound 2, which are
derivatives of benzimidazolium bromide, on human prostate cancer cell line (LNCaP)
concentrations of 10, 25, 50, 75, 100, 150, 200, 250 and 500 µM were investigated.
Furthermore, the time and dose dependent cytotoxic effects of 3- (4,5-DimethylThiazol-2-
yl) -2,5 diphenyltetrazolium bromide (MTT) test were investigated. 5-Fluoruracil (5-FU)
used as positive control and Compound 1 concentrations determined to be more cytotoxic
than MTT results were applied to LNCaP cells in a 1: 1 ratio and their antagonistic and
synergistic effects on cell proliferation were investigated. Finally, Apoptotic DNA
Fragmentation was determined using ELISA method.
Compound 1 was observed to have an effect on cell morphology at a concentration
of 100 µM in 24 hours and 25 µM in 48 hours. IC50 values were calculated as 183 and 90
µM for 24 and 48 hours, respectively. It was observed that Compound 2 began to affect cell
morphology at a concentration of 50 µM at 24 and 48 hours. IC50 values were calculated as
488 and 239 µM for 24 and 48 hours, respectively. In the doses of 5-FU, which was used as
a positive control, IC50 values were not found at 24 and 48 hours. In combined administration
(Compound 1 + 5-FU), antagonistic effect was observed after a dose of 200 µM in 24 hours
and 100 µM in 48 hours. When the Apoptotic DNA Fragmentation of 100 and 250 µM
concentrations of Compound 1 was investigated in the LNCaP cells, it was observed that the
necrotic effect did not occur, whereas the apoptotic effect increased depending on the
concentration cell death occurred with apoptosis
