Bu tez çalışması ile şap virüsüne duyarlı, şap hastalığının teşhisini ve şap virüsünün
izolasyonunu daha etkin kılacak alternatif yeni hücre hatları oluşturulması hedeflenmiştir.
Böylece, hem bu hücre hatlarının hastalığın doğal konakçısı ile aynı homolojiye sahip
olması hem de ilk enfeksiyon belirtilerinin görüldüğü ve enfeksiyona duyarlı bir organ olan
dilden hazırlanması nedeniyle şap virüsünün teşhisine ve izolasyonuna daha etkin katkı
sağlayacağı düşünülmüştür. Çalışmanın birinci aşamasında, mezbahadan alınan yaklaşık 5
aylık sığır fötüsü dilinden, güvenli laboratuvar koşullarında hazırlanan primer kültürün
ayrıştırma ve saflaştırılmasıyla, sonlu epitelyal ve fibroblast hücre hatları oluşturulmuştur.
İkinci aşamasında, oluşturulan bu iki yeni hücre hattının karakterizasyonları kapsamında,
mikrobiyal ve viral sterilite, büyüme karakteristiği, tür orijini doğrulaması, doku orijini
doğrulaması testleri ve şap virüsü için spesifik hücresel reseptörlerin varlığının tespiti
yapılarak tanımlı hücre hatları haline getirilmişlerdir. Üçüncü aşamasında ise yeni hücre
hatlarının şap virüsü laboratuvar suşlarına ve (A TUR/11, O TUR/07 ve Asia1/11) ve BHK
21 An31 hücre hattında negatif olan 20 adet saha suşuna olan duyarlılıkları incelenmiştir.
Çalışmalar sonucunda, FBTEC hücre hattının ömrünün 10 pasaj olduğu, FBTFC hücre
hattının ise 60. pasaja kadar çoğaldığı görülmüştür. Yapılan virüs duyarlılık testlerinde, her
iki yeni hücre hattının da kontrol grubu olan BHK 21 An31 hücre hattına göre ya eşdeğer ya
da daha duyarlı olduğu görülmüştür. Ancak FBTEC hücre hattının ömrünün çok sınırlı
olması bu hücre hattının uzun süreli kullanılabilirliğini sınırlamaktadır. FBTFC hücre hattı
ise hem uzun süre pasajlanabilir olması hem de diploit hücre özelliğiyle atasal özelliklerini
korumasından dolayı, iyi bir bankalama stratejisi ile daha uzun yıllar kullanılabilir yeni ve
alternatif bir hücre hattı olmuştur
In this thesis, new cell lines susceptible to foot and mouth disease virus (FMDV)
development, for diagnosis and isolation was aimed. These cell lines were prepared from the
natural host’s tongue where the first typical symptoms appear. In the first stage of the study,
fibroblastic (FBTFC) and epithelial (FBTFC) finite cell lines were formed from a 5 months
old bovine fetus’s tongue in accordance with safe laboratory conditions. In the second stage,
sterility, growth characteristics, purity, species and tissue origin verification, specific FMDV
receptors determination were done within the scope of the new cell lines’ characterization.
In the third stage, susceptibility to laboratory strains were examined in terms of antigenicity infectivity titers, infectivity period and plaque characters. 20 field strains, FMDV positive
with PCR but negative in BHK 21 An31 cells were tested with the new cells. CPE detection
and serotyping for FMDV were done after 3 blind passages and if any, performed by qRT PCR. The results revealed that degenerations and proliferation defects started in the FBTEC
cell line after the 10th passage in parallel with prolonged doubling time. FBTFC cell line
reached to the 60th passage with no morphological and proliferation problem. Both novel
cell lines were either equivalent or more sensitive than BHK 21 An₃₁ to FMDV. However,
short lifespan of the FBTEC cell line limits its long-term usability. The FBTFC is a new cell
line which can be used for years with a good banking strategy because of its longer passage
life and maintaining its ancestral properties with diploid cell feature
