Prostat kanseri erkeklerde akciğer kanserinden sonra en sık görülen kanser türüdür.
Bu sebeple yeni ve daha etkili tedavi seçenekleri arayışı her geçen gün artmaktadır. Yapılan
son çalışmalar benzimidazolyum türevlerinin kanser hücreleri üzerindeki inhibe edici
etkisini ortaya koymaktadır.
Çalışmada prostat kanseri hücreleri (DU145) kullanılarak yeni sentezlenen
benzimidazolyum türevlerinin (Bileşik A ve B) sitotoksik etkisi MTT yöntemiyle
araştırılmıştır. Bileşik A için 24 saatlik IC50 değeri 113 µM, 48 saat için ise 80 µM olarak
hesaplanmıştır. Bileşik B’nin 24 saat için IC50 değeri 420 µM, 48 saat için 363 µM olarak
belirlenmiştir. Bileşik A ve B’nin sağlıklı hücrelerde oluşturduğu sitotoksik etkiyi
belirlemek için insan sağlıklı akciğer bronşiyal epitelyal hücre hattı (BEAS-2B)
kullanılmıştır. Bileşik A’nın IC50 değeri 24 saatte 144 µM, 48 saatte ise 109 µM olarak
hesaplanmıştır. Bileşik B için 24 saatte hesaplanan IC50 değeri 307 µM, 48 saatte ise 200 µM
olarak belirlenmiştir. 5-Fluorourasil çalışmada pozitif kontrol olarak kullanılmış ve hem 24
hem de 48 saatlik uygulamalarında herhangi bir IC50 değeri hesaplanamamıştır. Bileşik A
konsantrasyonlarının 5-Fluorourasil dozları ile kombine uygulanması sonucunda 24 saatlik
IC50 değeri 91 µM, 48 saatlik uygulamada ise 66 µM olarak belirlenmiştir. DU145
hücrelerinde Bileşik A’nın 100, 150 ve 200 µM konsantrasyonlarının hem apoptotik hem de
nekrotik hücre ölümünü arttırdığı belirlenmiştir.
Sonuç olarak, DU145 ve BEAS-2B hücrelerine uygulanan Bileşik A ve B’nin doza
ve zamana bağlı olarak hücre çoğalmasını inhibe ettiği gözlenmiştir. Ayrıca, DU145
hücrelerinde Bileşik A konsantrasyonlarının 5-Fluorourasil dozları ile kombine
uygulanmasının sinerjistik bir etkiye neden olduğu belirlenerek Bileşik A’nın apoptotik ve
nekrotik hücre ölümünü arttırdığı tespit edilmiştir
Prostate cancer is the most common type of cancer in men after lung cancer. For this
reason, the search for new and more effective treatment options is increasing every day.
Recent studies reveal the inhibitory effect of benzimidazolium derivatives on cancer cells.
In the study, the cytotoxic effect of newly synthesized benzimidazolium derivatives
(compounds A and B) using prostate cancer cells (DU145) was investigated by MTT
method. The 24 hours IC50 value for compound A was calculated as 113 µM and 80 µM for
48 hours. The IC50 value of compound B was 420 µM for 24 hours and 363 µM for 48 hours.
Human healthy lung bronchial epithelial cell line (BEAS-2B) was used to determine the
cytotoxic effect of compounds A and B in healthy cells. The IC50 value of compound A was
calculated as 144 µM in 24 hours and 109 µM in 48 hours. For compound B, the IC50 value
calculated at 24 hours was 307 µM and at 48 hours it was 200 µM. 5-Fluorouracil was used
as a positive control in the study and no IC50 value was calculated in both 24 and 48 hours
applications. Combined administration of compound A concentrations with 5-Fluorouracil
doses determined that the 24 hours IC50 value was 91 µM and 66 µM in 48 hours application.
Concentrations of compound a of 100, 150, and 200 µM in DU145 cells have been found to
increase both apoptotic and necrotic cell death.
As a result, compounds A and B applied to DU145 and BEAS-2B cells have been
observed to inhibit cell proliferation depending on dose and time. Furthermore, combined
administration of compound a concentrations in DU145 cells with 5-Fluorouracil doses was
determined to cause a synergistic effect, and compound A was found to increase apoptotic
and necrotic cell death
