Nörodejenerasyonla ilişkili nöronal işlev bozukluklarını araştırmak için
kullanılan sinaptozomlar, sinaptik fonksiyonların incelenmesine olanak
sağlayan bir in vitro model sistemidir. Sirtuin 1 (SIRT1) ve poli(ADP-riboz)
polimeraz-1 (PARP1)’in yanı sıra peroksizom proliferatör ile aktive edilmiş
reseptör gama (PPARγ), Alzheimer hastalığında (AD) rol oynayan stres yanıtı
ve apoptotik yolakları regüle ederek nöroprotektif etkiler göstermektedir. AD
etyopatogenezinde henüz etkin bir tedavi şekli bulunmamaktadır. Bu nedenle,
potansiyel yeni terapötik ajanların AD’deki rollerini daha iyi anlayabilmek
için nöroprotektif sinyal yolakları üzerindeki etkilerinin araştırılması
gerekmektedir. Çalışmamızda, nükleer reseptör agonisti olan bexarotene ve
hücresel biyoenerjetiğin homeoastazından sorumlu olan nikotinamid, Aβ(1-42)
ile inkübe edilmiş sinaptozomlardaki nöroprotektif etkilerinin antioksidan,
oksidatif stres, apoptoz ve SIRT1/PARP1/PPARγ sinyal yolakları üzerinden
araştırılması amaçlanmıştır.
Çalışmamız in vivo ve in vitro olmak üzere 2 kısımdan oluşmaktadır. İn
vivo kısmında, her grupta altı hayvan olacak şekilde yirmi dört adet Wistar
albino erkek sıçan 4 gruba (kontrol, DMSO, nikotinamid ve bexarotene)
ayrıldı. Deney gruplarındaki hayvanlara 7 gün boyunca intraperitoneal olarak
DMSO (%1), nikotinamid (100mg/kg) ve bexarotene (0.1mg/kg) uygunlandı.
Ardından, hayvanlar dekapite edilerek serum ve beyin dokuları alındı.
Çalışmamızın in vitro kısmında ise, AD modelinin oluşturulmasında
kullanılacak olan sinaptozomların beyin dokusundan elde edilme için 3 farklı
izolasyon yöntemi (Whittaker, Percoll gradient ve sükroz gradient) kullanıldı.
Elektron mikroskop analiz sonuçlarına göre, en efektif izolasyon yönteminin
sükroz gradient prosedürü olduğu belirlendi. 10µM Aβ(1-42) konsantrasyonu
ile inkübe edilen sinaptozomlardaki TAS, TOS, CASP3, Cyt c, SIRT1, PPARγ
ve PARP-1 seviyeleri ELISA yöntemiyle ölçüldü.
Serum ve beyin örneklerindeki biyokimyasal analizler ve histopatolojik
incelemeler DMSO, nikotinamid ve bexarotene uygulamalarının sıçan beyin
dokusunda herhangi bir hasara neden olmadığını gösterdi. İn vitro Aβ(1-42)

uygulamasının sinaptozomlarda TAS, SIRT1 ve PPARγ seviyelerini
azaltırken, TOS, CASP3, Cyt c, ve PARP1 seviyelerini arttığını bulduk.
Nikotinamid tedavisinin antioksidan kapasitesini destekleyerek oksidatif
stresi ve apoptozu baskıladığı ve SIRT1 aktivasyonu üzerinden PPARγ
artırırken, PARP1 düşüşüne neden olmuştur. Bexarotene ise, PPARγ
aktivasyonu ile SIRT1 seviyelerinde ılımlı bir artışa neden olmuştur.
Sonuç olarak, AD patogenezinde nikotinamid sinaptozomlardaki
miktokondriyal fonksiyonları düzenleyerek bexarotene göre daha etkili
olabileceğini bulduk
Synaptosomes used to investigate neuronal dysfunctions associated with
neurodegeneration are an in vitro model system that allows examination of
synaptic functions. Sirtuin1 (SIRT1) and poly(ADP-ribose) polymerase-1
(PARP1) as well as peroxisome proliferator-activated receptor gamma (PPARγ)
have neuroprotective effects by regulating stress response and apoptotic
pathways involved in the Alzheimer's disease (AD). There is no effective
treatment for AD pathogenesis yet. Therefore, it is necessary to investigate the
effects of potential new therapeutics on neuroprotective signaling pathways in
order to understand their role in AD. In our study, we were aimed to
investigate the neuroprotective effects of bexarotene, nuclear receptor agonist,
and nicotinamide, responsible for the homeoastasis of cellular bioenesthetics,
in synaptosomes incubated with Aβ(1-42) through antioxidant, oxidative
stress, apoptosis and SIRT1/PARP1/PPARγ signal pathways.
Our study consists of 2 parts, in vivo and in vitro. In the in vivo section,
twenty four Wistar albino male rats were divided into 4 groups (control,
DMSO, nicotinamide and bexarotene) with six animals in each group. DMSO
(1%), nicotinamide (100mg/kg) and bexarotene (0.1mg/kg) were administered
intraperitoneally to animals in the experimental groups for 7 days. Then,
animals were decapitated and serum and brain tissues were removed. In the in
vitro part of our study, 3 different isolation methods (Whittaker, Percoll
gradient and sucrose gradient) were used to obtain the synaptosomes from the
brain tissue that will be used to create the AD model. According to the electron
microscope analysis results, the most effective isolation method was
determined to be the sucrose gradient procedure. TAS, TOS, CASP3, Cyt c,
SIRT1, PPARγ and PARP-1 levels in the synaptosomes incubated with a
concentration of 10µM Aβ (1-42) were measured by ELISA method.
Biochemical analysis and histopathological examinations in serum and
brain samples showed that DMSO, nicotinamide and bexarotene treatments
did not cause any damage to the rat brain tissue. We found that in vitro Aβ (1-
42) administration decreased TAS, SIRT1 and PPARγ levels in synaptosomes
while increasing TOS, CASP3, Cyt c, and PARP1 levels. Nicotinamide

treatment suppressed oxidative stress and apoptosis by supporting antioxidant
capacity and increased PPARγ through SIRT1 activation, causing PARP1
decrease. On the other hand, bexarotene caused a moderate increase in SIRT1
levels with PPARγ activation.
Consequently, we found that nicotinamide can be more effective than
bexarotene in AD pathogenesis by regulating mycochondrial functions in
synaptosomes
