Bu çalışmada, Cd ile kirlenmiş toprakların iyileştirilmesi için fitoekstraksiyon
yönteminin ve nanomateryallerin birlikte kullanılması ve aynı zamanda nanomateryallerin
bitkiler üzerindeki etkilerinin araştırılması amaçlanmıştır. Bu amaçla ülkemizde kullanımı
giderek yaygınlaşan, kuraklığa ve sıcaklığa dayanıklı, bol yeşil aksama sahip, ülkemizin
hemen hemen her yerinde yetiştirilebilen tütün bitkisi bitki materyali olarak kullanılmıştır.
Bitkiler, artan dozlarda ZnO-NP (0-1-5 mg/L) ve Cd (0-1-3 mg/L) uygulanmış Hoagland
besin çözeltisi ortamında 20 gün boyunca kontrollü koşullarda (16/8 saat ışık/karanlıkta,
25/20 oC sıcaklık ve % 60 nem, ışık yoğunluğu 10 klux) yetiştirilmiştir. Bitkilerin
uygulamalara tepkileri gözlemlenmiş ve analiz sonuçları değerlendirilmiştir. Deneme
sonunda yaprakların klorofil içeriği SPAD metre ile ölçüldükten sonra bitkiler hasat
edilmiştir. Bitki örneklerinin yeşil aksam ve kök kuru ağırlıkları ve çinko (Zn), kadmiyum
(Cd), azot (N), fosfor (P), potasyum (K), demir (Fe), bakır (Cu) ve mangan (Mn)
konsantrasyonları belirlenmiştir.
Çinko oksit NP uygulamalarındaki artışla yeşil aksam ve kök Zn konsantrasyonu
artmıştır. Çinko konsantrasyonu yeşil aksamda 17,84-3,360 mg/kg arasında ve kökte ise
39,74-8,80 mg/kg arasında değişim gösterdiği belirlenmiştir.
Artan Cd uygulamaları bitkilerin kök ve yeşil aksamlarındaki Cd birikimini
arttırmıştır. Diğer yandan ZnO-NP uygulamaları yeşil aksam Cd alımlarını azalmıştır.
Yeşil aksamda en yüksek Cd konsantrasyonu (388 mg Cd/kg) ZnO-NP uygulamasının 1
mg/L dozu ile 3 mg Cd/L dozlarının birlikte uygulamasından elde edilmiştir.
Yapılan tez çalışmasında, ZnO-NP uygulamalarının yaprakların klorofil içeriğini ve
biomas üretimini arttırarak bitkinin fitoekstraksiyon potansiyeline olumlu etkide
bulunduğu saptanmıştır
The aim of this study is to use phytoextraction method and nanomaterials together
in order to improve Cd contaminated soils, as well as investigating the effects of
nanomaterials on plants. For this purpose, the tobacco plant, whose usage has become
widespread in our country, and which is resistant to drought and temperature, has abundant
green parts and can be grown almost everywhere in our country, has been used as plant
material. Plants were grown in Hoagland nutrient medium with increasing doses of ZnO NP (0-1-5 mg/L) and Cd (0-1-3 mg/L) under controlled conditions (16/8 hours light/dark,
25/20 oC temperature and 60 % humidity, light intensity 10 klux) for 20 days. The
response of the plants to the applications was observed and the results of the analysis were
evaluated.
Plants were harvested, at the end of the experiment, after measurement of
chlorophyll content of leaves by using SPAD meter. Dry weights of shoot and root parts,
and the concentrations of zinc (Zn), cadmium (Cd), nitrogen (N), phosphorus (P),
potassium (K), iron (Fe), copper (Cu) and manganese (Mn) were determined in all plant
samples. The Zn concentration of shoots were determined varies between 17.84-3.360 mg/
kg. As for the root, it was found to vary between 39.74-8.80 mg/kg. The Zn concentrations
of shoot and the root were increased with the increase dose of ZnO NP applications.
The increasing dose of Cd applications were increased Cd accumulation in the root
and shoot parts of plants. On the other hand, Cd uptake of the shoot was decreased with
ZnO-NP applications. The highest Cd concentration (388 mg/kg) in the shoot was obtained
from 1 mg ZnO-NP/L and 3 mg Cd/L doses together.
In the thesis study, it was determined that ZnO-NP applications had a positive
effect on the phytoextraction potential of the plant by increasing the chlorophyll content
and biomass production of the leaves
