Çalışmamızda beyinde iskemi/reperfüzyon ile
oluşturulan hasar sonrası uygulanan A vitaminin morfolojik ve antioksidan etkilerini
araştırdık. Çalışmamızda 40 Sprague- Dawley sıçan her bir grup 8’er hayvan olacak
şekilde 5 gruba ayırdık. Kontrol grubu hariç tüm gruplarda sağ a. carotis communise
30 dakikalık iskemi ve bunu takiben reperfüzyon uyguladık. 24 saatlik iyileşme
sürecinin ardından 7 gün süreyle 3. gruba 1.000 IU/kg/gün, 4. gruba 3.000 IU/kg/gün,
dozunda 5. gruba ise 9.000 IU/kg/gün dozunda oral gavaj yoluyla A vitamini
uygulaması yaptık. 2. gruba iskemi/reperfüzyon uygulamasından sonra 7 gün boyunca
oral gavaj yoluyla ayçiçek yağı verdik. Kontrol grubuna ise normal değerleri elde
etmek amacı ile hiçbir uygulama yapmadık, sıçanlar standart pelet yem ile ad libitum
beslendi. 7 günlük uygulamanın sonunda intrakardiyak kan örnekleri ve sağ beyni
aldık. Kan örneklerinden süperoksit dismutaz (SOD), malondialdehit (MDA) ve
katalaz (KAT) seviyeleri belirlendi. Doku örnekleri ise hematoksilen/eozin ile
histokimyasal olarak boyandı ve beyin histolojisindeki değişiklikler ışık
mikroskobunda incelendi. Çalışma sonucunda A vitamini ile tedavi edilen gruplarda
iskemi/reperfüzyon grubuna göre beyinde oluşan hasar azaldı. SOD, MDA ve KAT
değerlerinin ise A vitamini uygulanan gruplarda kontrol grubu değerlerine göre
anlamlı olarak olumlu etkileri görüldü. Sonuç olarak, beyinde iskemi/reperfüzyon
sonrasında yapılan A vitamini uygulaması histolojik hasarı azaltıcı ve antioksidan
değerleri tedavi edici etki ortaya çıkarmıştır
In our study, we investigated the morphological and antioxidant
effects of vitamin A applied after injury caused by ischemia / reperfusion in the brain.
In our study, we divided 40 Sprague-Dawley rats into 5 groups, each group being 8
animals. In all groups except the control group, we applied 30 minutes ischemia to the
right a.carotis communis followed by reperfusion. Following the 24-hour recovery
period, we applied vitamin A via oral gavage for 7 days to the 3rd group at a dose of
1.000 IU/ kg/day, to the 4th group at a dose of 3.000 IU/kg/day, and to the 5th group
at a dose of 9.000 IU/kg/day. After the ischemia / reperfusion application to the second
group, we gave sunflower oil via oral gavage for 7 days. We did not apply any
application to the control group in order to obtain normal values, rats were fed ad
libitum with standard pellet feed. We received intracardiac blood samples and the right
brain at the end of the 7-day application. Superoxide dismutase (SOD),
malondialdehyde (MDA) and catalase (CAT) levels were determined from blood
samples. Tissue samples were histochemically stained with hematoxylin / eosin and
changes in brain histology were examined under a light microscope. As a result of the
study, brain damage decreased in the groups treated with vitamin A compared to the
ischemia / reperfusion group. SOD, MDA and KAT values were significantly positive
compared to the control group values in the groups treated with vitamin A. As a result,
application of vitamin A after ischemia / reperfusion in the brain has reduced
histological damage and therapeutic effect of antioxidant values
