Nöroblastom sempatik sinir sisteminden kaynaklanan bebeklik
çağında en sık görülen solid tümördür. Gelişimini ve farklılaşmasını tamamlamamış
nöral krest prekürsör hücrelerinden kaynaklandığı düşünülmektedir. Oleuropein
antioksidan, antienflamatuvar, antitümöral, antidiyabetik, antiaterojenik,
antihipertansif, hipolipidemik bir maddedir. Ayrıca hepatoprotektif, neuroprotektif
ve kardiyoprotektif etkisi de vardır. Yapılan deneylerimizde nöroblastoma
hücrelerinde Oleuropein’in nöroblastom hücreleri üzerindeki etkilerini
inceledik.Hücre kültürü çalışmamızda SHSY-5Y hücre hattını kullandık. DMEM
high glukoz ve F12 besiyerlerinin karışımı içine %10 FBS ve %1 penisilin streptomisin eklenerek elde edilen besiyerinde bu hücreler çoğaltıldı. Oleuropeinin
206 mM stok solüsyonu hazırlandı. Oleuropein’in bu hücreler üzerine etkisini
sitotoksisite testleri (MTT, Nötral red, Sulforhadamine B), ışık mikroskop
incelemeleri (Hematoksilen-Eozin boyama, immünhistokimyasal boyama), inverted
mikroskop incelemeleri, yara iyileşmesi testi ve western blot analizleri ile
araştırdık.Yaptığımız deneylerde Oleuropein’in artan dozlarında hücre canlılığının
azaldığı, çekirdek ve sitoplazma morfolojisinin bozulduğu, apoptotik yolaklarda
görev alan proteinlerin ifadesinin arttığı ve anti-apoptotik olanın ise azaldığı tespit
edildi.Sonuç olarak, Oleuropein’in nöroblastom hücreleri üzerindeki etkileri
nedeniyle nöroblastom tedavisi açısından incelenmesi gerektiğini düşünüyoruz
Neuroblastoma is the most common solid tumor originating from
the sympathetic nervous system. It is thought to be caused by neural crest precursor
cells that have not completed development and differentiation. Oleuropein is an
antioxidant, anti-inflammatory, antitumoral, antidiabetic, antiatherogenic,
antihypertensive, hypolipidemic substance. It also has hepatoprotective,
neuroprotective and cardioprotective effects. In our experiments, we examined the
effects of Oleuropein on neuroblastoma cells. We used SHSY-5Y cell line in our cell
culture study. These cells were grown in the medium obtained by adding 10% FBS
and 1% penicillin-streptomycin into a mixture of DMEM high glucose and F12
media. 206 mM stock solution of Oleuropein was prepared. We investigated the
effect of oleuropein on these cells with cytotoxicity tests (MTT, Neutral red,
Sulforhadamine B), light microscope examinations (Hematoxylin & Eosin staining,
immunhistochemical staining), inverted microscope examinations, wound healing
test and western blot analysis. In our experiments, it was determined that the cell
viability decreased in the increasing doses of Oleuropein, the nucleus and cytoplasm
morphology deteriorated, the expression of the proteins involved in the apoptotic
pathways increased and the anti-apoptotic one decreased In conclusion, we think that
Oleuropein should be examined in terms of neuroblastoma treatment due to its
effectsonneuroblastomacells
